Analysis of Phytochemicals in Herbal Extracts

Deionized water was used for HPLC-Q-TOF-MS and HPLC-Q-TOF-MS/MS analysis and was purified using a Milli-Q system (Merck Millipore, Billerica, MA, USA). HPLC-grade acetonitrile and formic acid were purchased from Merck (Darmstadt, Germany) and ROE (Newark, New Castle, USA), respectively. Methanol (AR) was purchased from the National Institutes for Food and Drug Control (Beijing, China). The seventeen reference substances (seven flavonol glycosides, three flavonols, two polymethoxyflavonoids, three coumarins, one alkaloid, and one limonin), including synephrine (15), neoeriocitrin (36), poncirin (39), eriocitrin (40), naringin (42), naringenin (43), narirutin (44), neohesperidin (51), hesperidin (56), apigenin (59), hesperitin (61), xanthotoxol (71), scopoletin (76), auraptene (80), limonin (79), nobiletin (85), and tangeretin (87) were purchased from Shanghai Yuanye Bio-Technology Co., Ltd. (Shanghai, China).

Quantitative Analysis of Bioactive Compounds

The standard reference materials of amygdalin, narirutin, neohesperidin and digoxin were purchased from Yuanye Bio-Technology Co., Ltd (Shanghai, China). digoxin is not the endogenous compound of XFZYD and plasma, and it does not obviously interfere with the retention times of all three analytes. Qualitative analysis was carried out using an LC-MS system (Shimadzu 8050, Kyoto, Japan) in negative ion mode. The plasma samples were added with digoxin, and then vortex mixed for 1 min and centrifuged for 15 min (13,000 rpm, 4°C) after being added with acetonitrile. The obtained supernatants were dried in a nitrogen dryer, diluted with 10% acetonitrile-water, repeated the extraction above, and injected into the LC-MS for analysis.

Quantification of Bioactive Compounds in Plasma

We purchased amygdalin, neohesperidin, rutin, and Digoxin from Yuanye Bio-Technology Co., Ltd. (Shanghai, China). Digoxin is a reference compound because it is not the internal composition of XFZYD and plasma; Digoxin does not disturb the residence times of the three objects. HPLC-MS/MS system (Shimadzu 8,050, Kyoto, Japan) was used for qualitative analysis in negative ion mode. After adding acetonitrile, mixing Digaoxin with the plasma samples, vortex the mixture (1 min), and centrifuge the mixture (13,000 rpm, 15 min, 4°C). Using a nitrogen dryer to dry the supernatants, diluted the dried supernatants (10% acetonitrile-water) and injected them into the HPLC-MS/MS for detection.

Profiling Aurantii fructus Metabolites

The LC–MS-grade acetonitrile and methanol were purchased from Merck (Germany).

Analytical Method Development for Herbal Compounds

Acetonitrile (ACN, LC-MS/MS grade) and formic acid (≥98%, analytical grade) were purchased from Fisher Scientific (Fair Lawn, New Jersey, USA) and Sinopharm Chemical Reagent Co., Ltd. The samples were identified by Dr. Sen Li of Huazhong University of Science and Technology.

Authentication of Herbal Medicines in MZRW

All the six herbal medicines (HMR: RM130121-08; DH: 130524; BS: RM130128-11; KXR: RM121026-04; HP: RM121026-05; ZS: RM121210-11) contained in MZRW were supported and authenticated by PuraPharm Company, Nanning department (Nanning, China). The reference standards of gallic acid, amygdalin, paeoniflorin, hesperidin, neohesperidin, naringin, magnolol, honokiol, aloe emodin, physcion, chrysophanol were purchased from Shanghai Yuan-Ye Bio-technology company (Shanghai, China). Emodin, rhein, albiflorin were purchased from the National Institute for the Control of Pharmaceutical and Biological Products (Beijing, China). The purity of all these reference standards was ≥98.0% (HPLC). HPLC grade of acetonitrile, methanol and formic acid were purchased from Merck (Darmstadt, Germany). Deionized water was purified by the Millipore water purification system (Millipore, Milford, MA, United States). All other reagents used were of analytical grade. Twelve male Sprague-Dawley rats (220–250 g) were kindly provided by the Experimental Animal Center of Chinese University of Hong Kong (Hong Kong). The rats were bred in an environmentally controlled room (22 ± 2°C, relative humidity 50 ± 20%) with a natural light-dark cycle for 7 days before the experiment carried out. The animal study was carried out in accordance with the Guideline for Animal Experimentation of Hong Kong.

In addition, acetonitrile of HPLC grade, together with formic acid of LC-MS grade was attained from Fisher Scientific (Fair Lawn, NJ, USA), followed by attaining the purified water from Wahaha (Hangzhou, China).
