Histopathological Lung Examination in Mice

For histopatological examination, the lungs from the sacrificed mice were removed and were infused with 10% neutral buffered formalin, and then paraffin-embedded for sectioning. Five micron sections of each lung were cut and stained with Hematoxylin and Eosin for histopathology. Lung sections were examined using light microscopy and images were obtained using an Olympus DP21 digital camera system.

Lung Histopathology Post-Influenza Challenge

Lungs were harvested following influenza virus challenge at the time of euthanasia or, in the case of survival, at the end of the experiment on D14 post-challenge. Lungs were inflated with 10% formalin and submitted to the Biorepository and Pathology Core at ISMMS for tissue processing and paraffin embedding. Lung-tissue sections 5 μm in thickness were stained using H&E and evaluated by a veterinary pathologist who was blinded to the treatment groups, with the exception of the PBS vaccinated + challenge and PBS mock challenge group. Images were captured under 200× magnification using an Olympus B×43 with an Olympus DP21 Digital Camera system.
