Phenotypic Characterization of HLA-A2 Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Surface staining was performed to characterise target cells phenotypically to detect HLA-A2 expression using an (APC-conjugated anti-human HLA-A2 antibody (BioLegend, 343308, Clone: BB7.2). Phenotypic analysis was also performed to assess the success of HHDII gene transfection using an FITC-conjugated anti-human β2m antibody (Sigma-Aldrich, SAB4700012, Clone: β2M-01). Cells were incubated with PI immediately prior to running the samples. At the end of the assay, cells were re-suspended in Beckman Coulter Isoton II™ diluent and analysed immediately on the Beckman Coulter Gallios™ flow cytometer.

Sorting MHC Class I Surface Positive Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
For EpiScan screens, 30 μl of antibody (APC-conjugated anti-human HLA-A2 antibody, BioLegend, 343308 or APC-conjugated anti-human B2-microglobulin antibody, BioLegend, 316312) in a total volume of 1.5 ml was used per 10 million cells. Staining was conducted for 30 min at 4°C; cells were then washed in PBS prior to sorting. Sorting was performed on a Sony MA900 instrument. For EpiScan screen sorts, the small remaining library negative population was used to draw the MHC class I surface positive gate. Within that gate, four bins subdivided the positive population into equal populations. Examples can be found in Extended Data Fig. 4.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
