Drug-induced Autophagy Modulation

Quantitative Analysis of Tricyclic Antidepressants

Methanol, hexane, isoamyl alcohol and acetonitrile were purchased from Caledon (Ontario, Canada). All reagents and solutions were either HPLC or analytical grades. Imipramine 25 mg tablets (Batch No:555) manufactured by Lorestan pharmaceutical company (Iran), and Tofranil® 25 mg tablets (Batch No:8006) was from Novartis (Switzerland).

Chronic Clomipramine and Memantine Administration

clomipramine hydrochloride (clomipramine; Sigma-Aldrich, São Paulo, Brazil) and memantine hydrochloride (memantine; Sigma-Aldrich, São Paulo, Brazil) were used. All drugs were dissolved in the drinking water (memantine at 0.006 mg/ml and clomipramine at 0.06 mg/ml concentration). The solutions were changed every two days, and the amount of liquid consumed was determined by changes in the bottle weight. The total volume was divided by the number of animals in each cage, and an average of 4.9 ml/day/animal was stably observed throughout the experiments.

Vitamin B6 Modulation of Antidepressant Effects

Drugs utilized were as follows: Vitamin-B6, 100 mg/kg (pyridoxine HCl, 100 mg/ml, Caspian Tamin Industry, Iran) (6); clomipramine, 10 mg/kg (Sigma-Aldrich, Germany); venlafaxine, 16 mg/kg (Sigma-Aldrich, Germany); fluoxetine HCl, 20 mg/kg (gift from Pars Daru, Iran) (10). All drugs were diluted by normal saline and injections were adjusted for 10 mL/kg mice body weight. All drugs were administered i.p. 30 min before testing. Vitamin B6 was administered daily for 3, 6 or 9 days in order to evaluate the best timing. Each experimental group comprised of 8 mice as follows: control group, vitamin B6 groups (3, 6 or 9 days), vitamin B6 plus clomipramine, fluoxetine, venlafaxine or vehicle groups.

Quantification of Drugs in Biological Samples

Cotinine (COT), trans-3-hydroxycotinine (OH-COT), cotinine-d3 and trans-3-hydroxycotinine-d3 were obtained from LGC Standards (Molsheim, France). Caffeine, ibuprofen, methadone, morphine, pseudoephedrine, codeine, cocaine, clomipramine, propranolol and Δ-9-tetrahydrocannabinol analytical reference standards presented in Methanol were obtained from Cerilliant (Sigma-Aldrich, Saint-Quentin-Fallavier, France). Potassium dihydrogen phosphate and dipotassium hydrogen phosphate were obtained from Sigma-Aldrich (St Louis, MO, USA). Methanol, hexane, isopropanol were of analytical grade and were obtained from Merck (Darmstadt, Germany). Sterile water was obtained from Fresenius (Sevres, France). Ammonia solution 25% was purchased from VWR (Fontenay-sous-Bois, France). Hydrochloric acid 0.2 m was obtained from VWR (Fontenay-sous-Bois, France). Clean Screen SPE columns 200 mg were purchased from UCT (Bristol, PA, USA). The derivatizing agent utilized was bis(trimethylsilyl)trifluoroacetamide (BSTFA) + 1% trimethylchlorosilane (TMS) (Sigma-Aldrich, Saint-Quentin-Fallavier, France).

Antidepressants and Ketamine Behaviors

Pharmacological treatment of tumors

Pimozide (cat. no. P1793; Sigma-Aldrich) was intraperitoneally (IP) administered daily at 5 mg/kg. Risperidone (cat. no. 1604654; Sigma-Aldrich) was delivered IP daily (3 mg/kg) for 2 weeks (early lesion study) or in drinking water (1.56 mg/l) until euthanasia (tumor latency study), resulting in the same daily dose based on the calculation previously reported [37 (link)]. SML0935; Sigma-Aldrich) was delivered via IP injection in a daily dose of 3 mg/kg for 2 weeks, and clomipramine (cat. no. 1140247; Sigma-Aldrich) was delivered in drinking water (190 mg/l), resulting in a daily dose of 28 mg/kg. All drugs were diluted in dimethyl sulfoxide (DMSO) to the appropriate concentrations. Both ruxolitinib and control chow was provided by Incyte Corp. Ruxolitinib chow was packaged in a pre-determined measurement of 2000 mg/kg chow; mice were allowed to free-feed for the duration of the study.
