Culturing Murine and Human Cell Lines

L929 cell line derived from murine fibroblasts (ATCC CCL-1™) and U-2 OS cell line derived from human osteosarcoma (ATCC HTB-96™) were maintained in MEM (Minimal Essential Medium, Sigma, M0446) and DMEM (Dulbecco’s Modified Eagle’s Medium, Sigma, D0819), respectively with 10% FBS at standard conditions of 37 °C, 5% CO2 and 100% relative humidity. Cells were used from the 3rd passage after thawing and only till the 20th passage. Cells were passaged regularly using trypsin–EDTA solution (Sigma, T4049) which was stopped by addition of the medium with FBS with no subsequent centrifugation.

Generation of Stable GFP-AURKA Cell Lines

U2OS cells free from mycoplasma were purchased from American Type Culture Collection (ATCC, HTB-96) and were grown in Dulbecco's modified Eagle's medium (DMEM, Sigma-Aldrich) supplemented with 10% fetal bovine serum (Life Technologies, Thermo Fisher Scientific), 1% L-glutamine (Life Technologies, Thermo Fisher Scientific) and 1% penicillinstreptomycin (Life Technologies, Thermo Fisher Scientific). Cells were cultivated at 37°C and 5% CO2. The generation of GFP-AURKA and GFP-AURKA-mCherry stable cell lines were generated by transfecting U2OS cells with X-tremeGENE HP transfection reagent (Roche), following the manufacturer's indications. Stable clones were selected in DMEM supplemented with 10% fetal bovine serum, 1% L-glutamine, 1% penicillin-streptomycin and 500 μg/ml Geneticin (Invivogen). Stable and transient transfections were performed with X-tremeGENE HP transfection reagent (Roche) according to the manufacturer's instructions.

Culturing Glioblastoma, Astrocyte, and Osteosarcoma Cells

The T98G glioblastoma multiforme cell line (ATCC CRL-1690) was obtained from ATCC and cultured based on the specifications provided by ATCC. The human astrocyte cell line (ScienCell 1800) was purchased from ScienCell and was similarly cultured based upon the directions provided by ScienCell. GM00637 normal human fibroblasts, SV-40 transformed (Coriell) and U2OS human bone osteosarcoma epithelial cells (ATCC HTB-96) were cultured in DMEM (Sigma D5796) containing 10% FBS (Gemini 100-106) and 1% penicillin/streptomycin (Gibco 15070063).

Culturing U2OS Osteosarcoma Cells

The U2OS human bone osteosarcoma cell line (DSMZ, Braunschweig Germany) was obtained from ATCC (cat. HTB-96) and grown in Dulbecco's modified Eagle's medium nutrient mixture F-12 HAM (Sigma-Aldrich, St. Louis, MO) supplemented with 10% fetal bovine serum (Sigma-Aldrich), MEM nonessential amino acids (Sigma-Aldrich), and gentamicin (Sigma-Aldrich) at 37 °C in a humidified atmosphere supplemented with 5% CO 2 .
