Total cholesterol

Manufactured by EKF Diagnostics

The Total Cholesterol test is a laboratory analysis used to measure the total amount of cholesterol present in an individual's blood sample. This measurement includes both low-density lipoprotein (LDL) and high-density lipoprotein (HDL) cholesterol levels.

Biomarker Profiling in Fasted Plasma

Check if the same lab product or an alternative is used in the 5 most similar protocols
Plasma was collected at the indicated time points following a 4-hour fast. Insulin and adiponectin were quantified using commercially available ELISA kits (EMD Millipore). SAA (R&D), total cholesterol (Stanbio) and triglycerides (Stanbio) were measured using ELISA (SAA) and colorimetric assays, as described previously (den Hartigh et al., 2014 (link)). Fasting non-esterified fatty acid (NEFA) was measured using the NEFA-C (Wako) according to the manufacturer’s instructions.

Biochemical Analyses of Serum Markers

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood was collected by cardiac puncture into centrifuge tubes before centrifugation at 10,000 rpm for 15 min. Serum was separated and stored at −80°C. Serum FGF21 and insulin concentrations were measured according to the manufacturer’s instructions using quantitative ELISAs specific for mouse FGF21 and insulin (R&D Systems and Crystal Chem, respectively). ALT and AST (Pointe Scientific, Canton, MI), triglycerides, β-hydroxybutyrate, total cholesterol, glucose (StanBio, Boerne, TX) were measured in duplicate using enzyme colorimetric assays.

Serum Biomarker Profiling Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood was allowed to clot, and supernatant was collected after centrifugation at 4 °C. Serum FGF21 levels were determined using a Rat/Mouse FGF-21 ELISA kit (R&D Systems, Minneapolis MN), according to the manufacturer's instructions. α-Feto protein/AFP was measured using a mouse Quantikine ELISA Kit (R&D Systems, Minneapolis MN). Alanine aminotransferase (ALT) (Pointe Scientific, Canton MI), aspartate aminotransferase (AST) (Pointe Scientific, Canton MI), triglycerides, total cholesterol (StanBio Laboratory, Boerne, TX) and nonesterified fatty acids (Wako Diagnostics, Richmond, VA) were measured in duplicate using enzyme colorimetric assays as per the directions provided by each manufacturer.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
