Biomarker Profiling in Fasted Plasma

Plasma was collected at the indicated time points following a 4-hour fast. Insulin and adiponectin were quantified using commercially available ELISA kits (EMD Millipore). SAA (R&D), total cholesterol (Stanbio) and triglycerides (Stanbio) were measured using ELISA (SAA) and colorimetric assays, as described previously (den Hartigh et al., 2014 (link)). Biochemical Analyses of Serum Markers

Blood was collected by cardiac puncture into centrifuge tubes before centrifugation at 10,000 rpm for 15 min. Serum was separated and stored at −80°C. Serum FGF21 and insulin concentrations were measured according to the manufacturer’s instructions using quantitative ELISAs specific for mouse FGF21 and insulin (R&D Systems and Crystal Chem, respectively).
