Anticancer Potential of Baicalein

All reagents used for experimental purpose were of analytical reagent grade. Extra pure methanol, Baicalein, RuCl3. xH2O, DPPH (2, 2-diphenyl-2-picrylhydrazyl), ABTS (2, 2'-azinobis 3-ethylbenzothiazoline-6-sulphonic acid diammonium salt), TPTZ (2, 4, 6-Tri(2-Pyridyl)-s-triazine), highly polymerized CT-DNA (calf thymus DNA), Tris HCl, 7, dimethylhydrazine (DMH), dextral sodium sulphate (DSS), biotinylated goat anti-rabbit IgG, streptavidin peroxidase, 3,3'- diaminobenzidine (DAB), proteinase K, foetal bovine serum (FBS), insulin L-glutamine, sodium pyruvate, streptomycin, penicillin, MTT (3- (4,5-dimethyl thiazole-2-yl)-2,5-diphenyltetrazoliumbromide), Annexin V and propidium iodide (PI) were purchased from Sigma Aldrich Chemical Co (St, Louis, Mo, USA). Rabbit anti-rat p53, Bax, Bcl2, PCNA, VEGF, caspase-3, mTOR, WNT, Beta catenin were purchased from ANASPEC Inc. (San Jose, CA, USA). The HT-29 colorectal cancer cell line was procured from National Centre Cell Science (NCCS), Pune, India. Apoptosis detection kit obtained from Takara Bio Inc (Japan). Other reagents used for the experimental purpose were obtained in the purest form from local firms.

Histological and Immunohistochemical Analysis of Fish Samples

Fish samples were fixed in 10% neutral buffered formalin solution (Sigma, #HT5012). After dehydration and embedding with paraffin, sectioning was performed using a Leica microtome, and all the samples were sectioned sagittally at a thickness of 4 μm. For ordinary observation, the samples were stained with Hematoxylin (Sigma, #51275) and Eosin (Sigma, #HT110232). For immunohistochemistry, the primary antibodies used were PCNA (AnaSpec Inc. #55421), β-catenin (Abcam, #ab32572) and E-cadherin (Abcam, #ab1416). After washes with phosphate buffered saline (PBS) with 0.1% Tween 20 (PBST) and incubation with appropriate secondary antibody from the Dako Kit set (DAKO, Denmark, #346811), color was developed by using the Liquid DAB & Substrate Chromogen system (DAKO, #346811). The slides were also conter-stained with 5 μg/ml DAPI (4′, 6-Diamidine-2′-phenylindole dihydrochloride) to facilitate nucleus observation.

Antibody-based protein expression analysis

We used antibodies for β-actin (AC-15) and CPEB4 (HPA038394) from Sigma-Aldrich; PCNA (#53764) from AnaSpec; CCNB1 (#4138) from Cell Signaling Technology; and E-cadherin (sc-59905) from Santa Cruz Biotechnology. CPEB4 polyclonal and monoclonal antibodies raised against the N-terminal 427 amino acids (a.a.) of rat CPEB4 were as described [31 (link), 33 (link)]. With the exception of the Vectastain Elite ABC kit (cat No. PK-6102, Vector labs), all other chemicals were purchased from Sigma-Aldrich.
