All incubation steps were carried out under rotation at 4°C. GST or GST-tagged proteins were immobilized from E. coli extracts on 100 μl Glutathione–Sepharose beads in PBS (GE Healthcare). After incubation for 1 h at 4°C, with either E. coli extracts or purified proteins, the beads were washed three times, the bound material was eluted with GST elution buffer (50 mM Tris, 20 mM reduced glutathione), and subjected to SDS-PAGE followed by Coomassie Blue staining and western blot analysis using anti-His (H1029), or anti-GST (G1160) antibodies (Sigma-Aldrich).
