γ 32p atp 5 mci

Manufactured by PerkinElmer

[γ−32P]ATP (5 mCi) is a radioactively labeled nucleoside triphosphate used in various biochemical applications. It contains the radioactive isotope phosphorus-32 (32P) incorporated into the gamma-phosphate group of adenosine triphosphate (ATP). The specific activity is 5 millicuries per milligram.

Biochemical Characterization of PP1 Analogues

Check if the same lab product or an alternative is used in the 5 most similar protocols
All biochemicals, including bicine, LB, metal salts and antibiotics were purchased from either Melford or Sigma and were of analytical quality. [γ−32P]ATP (5 mCi) was purchased from Perkin-Elmer and diluted appropriately with unlabelled ATP. Sypro Orange dye was from Invitrogen and was diluted as described in the Invitrogen TSA protocol. A small compound library composed of 30 modified PP1 (pyrazolo-pyrimidine) analogues was prepared as previously described [45 (link)]. The compounds contain bulky additions at the C5 position of the pyrimidine ring that are designed to interact with the hydrophobic pocket adjacent to the gatekeeper residue of AS protein kinases. Compounds were stored at 10 mM in DMSO at −80°C until required and diluted directly into the assay at 250 μM, so that the final DMSO concentration was no higher than 2.5% (v/v) in any assay. Acronyms for compounds A, B and F are dClB-PP1 (A): dichlorobenzyl-PP1; dmB-PP1 (B): dimethylbenzyl-PP1; bisPh-PP1 (F): bisphenyl-PP1. Similar data were obtained from two independent compound preparations.

Radiolabeled Oligonucleotide Biochemical Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
All oligonucleotides used for the in vitro biochemical assays were purchased from Keck Biotechnology Resource Laboratory at Yale University and purified by denaturing polyacrylamide gel electrophoresis. [γ-32P]ATP (5 mCi) and ATP were purchased from PerkinElmer Life Sciences and Sigma, respectively. T4 polynucleotide kinase was purchased from New England Biolabs. 5-fluorouracil was purchased from Sigma. The ATR inhibitor VE-821 was purchased from Selleckchem.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
