Synthesis and Characterization of Iron Oxide Nanoparticles

Sodium oleate 82%, FeCl3·6H2O 97%, oleic acid 90%, octadecene 90%, dimercaptosuccinic acid 98% (DMSA), dimethyl sulfoxide ≥99.9% (DMSO), acetic acid, and 12 kDa cellulose membrane were purchased from Sigma-Aldrich (Madrid, Spain). Absolute ethanol 99.5%, hexane 95% and toluene 99.5% were purchased from PanReac (Barcelona, Spain). All other chemicals (NaOH, HNO3 65%, HCl 37%, NaCl, MgCl2, etc.) were of the highest quality commercially available and used as received. Milli-Q water (resistivity 18.2 MΩ·cm at 25 °C) was used in all experiments. Type-I collagen powders extracted from rat tails (N_COL) were provided by NOVAICOS Srl (Novara, Italy, https://www.novaicos.com/ (accessed on 20 December 2021)).

Synthesis of Water-Soluble CdSe Nanocrystals

Water-soluble CdSe core NCs were obtained using mercaptosuccinic acid as a surface-modifying agent. Briefly, 20–30 mg of NCs was dissolved in chloroform, after which 3 mL of dimercaptosuccinic acid (Sigma, dissolved in 150 mM PBS, pH 7.3) was added to 6 mL of NCs in chloroform. Water containing water-soluble CdSe NCs was carefully decanted and subjected to ultrafiltration to remove free (nonreacted) dimercaptosuccinic acid. The ultrafiltration was carried out using a concentrator (Vivaspin-6/20, Sartorius AG, Gottingen, Germany).

Phalloidin-Alexa 488 was acquired from Molecular Probes (A12379) and Fluoromount-G + DAPI from EMS (17984-24). Dulbecco's modified Eagle medium (DMEM) was purchased from Lonza (Ref. 12-614F). Dulbecco's phosphate buffered saline (DPBS 1×, Ref. 17-512F) and PBS (Ref. BE02-017F) were also purchased from Lonza.

High Wycombe, UK) was purchased from Shifa International Hospital Pharmacy (Islamabad, Pakistan). The ELISA kit was purchased by Nanjing Pars Biochem Ltd. (Nanjing, China) The kit contained standards, antibody-coated microtiter plate, chromogen A, chromogen B, HRP conjugate and stop solution.
Blood was collected from healthy donors in citrated and EDTA-containing vials. Vials and Aspirin (High noon Laboratories Ltd., Lahore, Pakistan) were purchased from a local pharmacy. LPS extracted from E. coli was a gift from the National Institute of Health, Islamabad, Pakistan. MCF-7 cell lines (HTB-22™) were purchased from ATCC (Manassas, VA, USA).
