Liposome-Mediated Delivery of Phospholipids

Phospholipid, 1,2-distearoyl-sn-glycero-3-phosphocholine (DSPC), 1,2-dioleoyl-sn-glycero-3-phosphoethanolamine-N-(carboxyfluorescein) (ammonium salt), L-α-phosphotidylglycerol sodium salt (FITC-PC) and PC 16:0/18:1 was commercially obtained (Avanti Polar Lipids, Inc., Alabaster, Al). Cholesterol was purchased from Calbiochem (San Diego, CA, USA). Liposome formulations were prepared by lipid film hydration and sonication methods as described previously (Mfuh et al. 2011 (link); Li et al. 2012a (link), b (link)). Liposome encapsulated PC 16:0/18:1 or liposomes alone were introduced into Hela and 104C1 cells in triplicate at a concentration of 25 µM/well. Following centrifugation at ~1000×g for 60 min at 25 °C, cells were washed with D-10 and infected with Ct D as described above.

PEth 16:0/18:1, PEth 16:0/18:2, PC 16:0/18:1, and PC 16:0/18:2 were obtained from Avanti Polar Lipids (Alabaster, USA). PLD and n-hexane were from Sigma-Aldrich (Buchs, Switzerland). Acetone, ammonium acetate, chloroform (CHCl 3 ), D 6 -ethanol, diethyl ether, CaCl 2 , and Na 2 HPO 4 were obtained from Merck (Darmstadt, Germany). 2-Propanol was provided by Fisher Scientific (Loughborough, UK), methanol (MeOH) was from Biosolve BV (Valkenswaard, the Netherlands), and acetonitrile (MeCN) was supplied by Agros Organics (New Jersey, USA). Deionized water was produced in-house with a Milli-Q water system from Millipore (Billerica, USA). Blank blood was obtained from the blood donation center in Bern, Switzerland.
