Quantification of Remdesivir and its Metabolite

Remdesivir was provided by Formosa Laboratories Inc. (Taoyuan, Taiwan). GS-441524 was purchased from Selleck Chemicals (Houston, TX, USA). Formic acid, polyethylene glycol 400 (PEG400), and urethane were obtained from Sigma-Aldrich (St. Louis, MO, USA). Critic acid, dextrose, sodium chloride, and sodium citrate were purchased from Merck (Darmstadt, Germany). MS-grade acetonitrile (ACN) was acquired from J.T. Baker Inc. (Phillipsburg, NJ, USA). Triply deionized water (Millipore, Bedford, MA, USA) was used to prepare the stock solutions and mobile phase. Standard stock solutions of remdesivir and GS-441524 were prepared using 10% DMSO in ACN, at a concentration of 1 mg/mL, and stored at -20°C. Calibration curves of remdesivir and GS-441524 were prepared using blank matrices (blood, fetus, placenta, and amniotic fluid) as 5 μL standard stock solutions added into (spike) 45 μL blank matrices, to generate solutions with concentrations of 0.5, 1, 5, 10, 50, and 100 ng/mL and 5, 10, 50, 100, 250, and 500 ng/mL, respectively.

In Vitro Cytochrome P450 Enzyme Assay

The following compounds were obtained from Alfa Aesar (Heysham, UK): 3′,4′,7,8-TMF (78-TMF),3′,4′,5,7-TMF (57-TMF), 3-hydroxy-2′,4′,5′,7-TMF (3H7-TMF), and 3-hydroxy-2′,4′,5′,6-TMF (3H6-TMF). P450 Baculosomes® (microsomes prepared from insect cells containing a single human P450 and P450 reductase), control Baculosomes® (microsomes prepared from insect cells infected with a control virus), regeneration system (glucose-6-phosphate dehydrogenase and glucose–6–phosphate), NADP+, Vivid® substrates, and assay buffers were obtained from Invitrogen Corp. (Madison, WI, USA). Corning provided 384 low–volume black polystyrene plates for the assays (Acton, MA, USA).

Sorafenib Quantification in H. diffusa

Pharmaceutical. (Taoyuan, Taiwan). The internal standard (diethylstilbestrol)
with a purity higher than 99% was obtained from Sigma-Aldrich Chemicals
(St. Louis, MO, USA). Other liquid chromatography grade reagents,
including methanol, acetonitrile, and potassium dihydrogen phosphate
monohydrate, were all obtained from E. Merck (Darmstadt, Germany).
All reagents used in the HPLC experiment were prepared with triply
deionized water (Millipore, Bedford, MA, USA).
Sorafenib and
diethylstilbestrol were dissolved in methanol and diluted with 50%
methanol to the specified concentrations as stock solutions. The calibration
and internal standard solution were stored at −20 °C for
subsequently analysis. Before drug administration for the animal experiment,
the H. diffusa extract was prepared
with triply deionized water.
