Quantitative PCR Protocol for MSCs and MNCs

Check if the same lab product or an alternative is used in the 5 most similar protocols
Total RNA was isolated from MSCs after five days of culture before seeding them on POMA slides for ECM generation as well as from 24h stimulated MNCs using RNeasy Mini Kit (Qiagen, Germany) and reverse transcribed into cDNA using RevertAid cDNA synthesis kit (Thermo-Fisher, Germany) with oligo-dT primers. Relative target quantity was determined using the comparative CT(ΔΔCT) method. RT-PCR was performed using SYBR Green/ROX PCR master mix (Thermo-Fisher, Germany) and target specific primers (Supplementary Table S2) on a 7500 Real-time PCR cycler (Applied Biosystems, Germany). Amplicons were normalized to U6, respectively. All samples were run in duplicates.

Quantifying Gene Expression in CD34+ HSPCs

Check if the same lab product or an alternative is used in the 5 most similar protocols
Total RNA were isolated from CD34+ HSPCs after incubation with EVs using RNeasy Micro Kit (Qiagen, Germany) and reverse transcribed into cDNA using RevertAid cDNA synthesis kit (Thermo Fisher Scientific, Germany) with oligo-dT primers. Relative target quantity was determined using the comparative CT (∆∆CT) method. RT-PCR was performed using SYBR Green/ROX PCR master mix (Thermo Fisher Scientific, Germany) and target specific primers for PTEN, CDKN2A and SIRT1 (Supplementary Table S4) on a Quantstudio three cycler (Applied Biosystems, Germany). Amplicons were normalized to endogenous GAPDH control. All samples were run in duplicates.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
