Preparation of ECM-Mimicking Lipid Vesicles

LUVs, mimicking the membrane domain involved in epithelial cell–ECM adhesion (Márquez et al. 2008 (link)), were prepared in pH 7.4 buffer containing 10.0 mM HEPES, 50.0 mM KCl, 1.0 mM EDTA, and 3.0 mM NaN3, as previously described (Su et al. 2019). Note that POPC, POPE, and SM have neutral charge, while POPS and POPI are negatively charged. Therefore, the ECM-adhesion-imitating LUVs used here are about 25% anionic. The size of the LUVs was 400 nm.

Liposome Preparation and Composition

Large unilamellar vesicles (LUVs), with a diameter of approximately 100 nm, were obtained by extrusion of multilamellar vesicles (MLVs) as described elsewhere [41 (link)]. 1-Palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC), 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphoethanolamine (POPE), 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphoinositol (POPI), and 1-palmitoyl-2-oleoyl-sn-glycero-3-phospho-L-serine (POPS) were from Avanti Polar Lipids (Alabaster, AL, USA), while cholesterol (Chol) and ergosterol (Erg) were from Sigma. The LUVs studied were mostly zwitterionic (pure POPC, POPC:Chol 70:30, POPC:Erg 70:30, and C. albicans membrane-like mixture, i.e., POPC:POPE:POPS:POPI:Erg 59:21:3:4:13 [42 (link)]), mimicking membrane systems relevant for the study [32 (link),41 (link)]. Liposomes were extruded in the day of the measurement. HEPES buffer was used in all the measurements, except when otherwise indicated.

Phospholipid Analysis Protocol

(Alabaster, AL, USA) and used without further purification. Milli-Q
water (Milli-Q H2O) was used for all the experiments, filtered
through a 0.22 μm filter, and obtained with Milli-Q Millipore
system (Synergy, Millipore Corporation, Billerica, MA, USA). HPLC
grade acetonitrile, methanol (MeOH), and chloroform (CHCl3) were obtained from Fisher Scientific Ltd. (Leicesterscire, UK).
Ammonium formate, formic acid, and solid nitronium tetrafluoroborate
(NO2BF4) were obtained from Sigma-Aldrich (St.
Louis, MO, USA). Sodium dihydrogen phosphate dihydrate (NaH2PO4·2H2O) and ammonium molybdate were
purchased from Piedel-de-Haën (Seelze, Germany). Ascorbic acid
and perchloric acid 70% were purchased from VWR International (Leicestershire,
UK) and from Panreac (Barcelona, Spain), respectively.
