Visualizing Differentiated Muscle Cells

The DPS with differentiated muscle cells were washed with PBS and fixed with 4% v/v PFA solution for 30 min. Fixed cells were treated with ethanol gradient concentration (30–100%) intended for dehydration and followingly dried the DPS with critical point drying (CPD) (Leica EM CPD300 Critical Point Dryer). Dried DPS were further coated with platinum (Pt) to visualise under SEM. JSC-1200 fine coater (JEOL, Japan) was used to sputter coat DPS for a thickness of 15 nm. Sputter-coated DPS were then analysed using FESEM (JSM-6701F, JEOL, USA). Morphological analysis of pADMSCs and C2C12 grown on the DPS were imaged using FESEM.

Nanofibrous Mats Morphology Characterization

Morphology of different nanofiber mats were examined using field-emission scanning electron microscope (FE-SEM; JEOL -JSM6701F, the Netherlands) and transmission electron microscope (TEM; JEOL JEM-3010).
For FE-SEM analysis, the samples were first sputter coated with platinum (JEOL JSC-1200 fine coater, Japan) and viewed at an accelerating voltage of 5kV. To estimate the average fiber diameter of various nanofibrous mats, ~50 distinct individual fibers from different areas were randomly selected and used for measuring their diameter using Image J software. In the TEM study, the fibers were collected on gold-coated copper specimen grids without staining. observed for various mats were then estimated to determine their antimicrobial efficiency.
