Characterization of Acetaminophen Formulations

HPLC grade methanol was from Thermo Fisher Scientific (Sunnyvale, CA, USA). Acetonitrile was from Global Science (Auckland, New Zealand). Potassium phosphate dibasic, potassium phosphate monobasic, Tween 80 and hydrochloric acid (37%) were from Merck KGaA (Darmstadt, Germany). Sodium phosphate tribasic dodecahydrate, acetaminophen and benzene-1,2-dithiol were from Sigma-Aldrich (St Louis, MO, USA). Polyethylene glycol 400 were from BDH laboratory reagents (Poole, England). DRcapsTM were purchased from Capsugel® (Morristown, NJ, USA), Eudragit® L100 and Eudragit® L100-55 powder was purchased from Evonik Australia Industries (Melbourne, Victoria, Australia), propylene glycol BP was purchased from PSM Health care (Auckland, New Zealand), and the hard gelatin capsules (size 0) were obtained from NZ Nutritionals® (Christchurch, New Zealand). All the other reagents and solvents were purchased from Sigma Aldrich (Auckland, New Zealand).

Photofunctionalisable Hydrogel Fabrication

Chemicals required to synthesise photolabile monomer included 4,5-dimethoxy-2-nitrobenzyl chloroformate (NVOC), methacrylamide poly(ethylene glycol) amine hydrochloride (Mn: 400), triethylamine (TEA), anhydrous tetrahydrofuran (THF), ethyl acetate, hydrochloric acid (HCl), and sodium sulphate (Na2SO4). These chemicals were bought from Sigma-Aldrich.
Ethanol, chloro(dimethyl)vinyl silane (CDVS), toluene, 40% (w/v) solution of 29 : 1 (w/w) acrylamide : bis-acrylamide, ammonium persulphate (APS), N,N,N′,N′-tetramethylethylenediamine (TEMED), sodium phosphate monobasic monohydrate, sodium phosphate dibasic dodecahydrate, sodium phosphate tribasic dodecahydrate, 1 M sodium hydroxide, hydroxylamine (NH2OH) hydrochloride, and fluorescein isothiocyanate (FITC) were bought from Sigma-Aldrich. Microscope glass slides were bought from VWR. These materials and chemicals were used to fabricate photofunctionalisable hydrogel films on glass substrates.
Materials used to make fluidic flow cells consisted of 3 mm thick transparent poly(methyl methacrylate) (PMMA) sheets, bootlace ferrules (211-4252), and nitrile O-rings (01-08-01801), and were purchased from Fred Aldous, RS Components, and Ashton Seals, respectively.

Formulation and Characterization of Tea Seed Oil-Based Microemulsions

The tea seed oil was obtained from the Tea Oil and Plant Oils Development Center, Chaipattana Foundation (Chiang Rai, Thailand). Soy lecithin fluid was obtained from Rama Production Co., Ltd. (Bangkok, Thailand). Tween®80, Span®80, Oil Red O and sodium phosphate tribasic dodecahydrate were purchased from Sigma-Aldrich Co. (St. Louis, MO, USA). Hydrochloric acid (37%), sodium chloride and sodium hydroxide were purchased from CARLO ERBA Reagents S.A.S. (Val de Reuil, France). Deionized water was used for the preparation of all the solutions.

Quercetin-Riboflavin Interaction in Whey Protein

The following chemicals were purchased from Sigma-Aldrich (St. Louis, MO, USA): quercetin (� 95 %, HPLC), riboflavin (� 98 %), sodium azide, ANS (8-Anilino-1-napththalenesulfonic), and sodium phosphate tribasic dodecahydrate 98 %. Whey Protein Isolate (WPI) powder (Lacprodan DI-9212), kindly supplied by Arla Foods Ingredients (Viby, Denmark), has in a total protein content of 91 % (in dry weight), moisture content of ca. 6 % and vestiges of lactose (max. 0.5 %) and fat (max. 0.2 %), according to the information provided by the supplier. Sodium hydroxide and phosphoric acid were obtained from Merck (Merck KGaA, Darmstadt, Germany). Hydrochloric acid and monosodium phosphate were purchased from Panreac (Barcelona, Spain), whereas sodium phosphate dibasic, acetonitrile (HPLC grade) and methanol were obtained from Chem-Lab (Zedelgem, Belgium). Dimethyl sulfoxide was obtained from Fisher Scientific, while ethanol was purchased from JMGS (Portugal). All other chemicals used in this study were reagents of analytical grade.

Nanoscale Hydroxyapatite Scaffold Coating

A nnHA coating was applied to the scaffolds as previously described (Eichholz et al., 2020a (link)). Briefly, the autoclaved samples were immersed in 70% ethanol at room temperature under vacuum to remove air bubbles. Scaffolds were then incubated with 2 M NaOH for 45 min at 37°C, rinsed in MilliQ water five times and then incubated with calcium solution (0.05 M Calcium Chloride Dihydrate (Sigma-Aldrich, United Kingdom) in MilliQ water). An equal volume of phosphorus solution, (0.03 M Sodium Phosphate Tribasic Dodecahydrate (Sigma-Aldrich, United Kingdom) in MilliQ water), was slowly added to the calcium solution. Air bubbles were removed under a vacuum, and samples were incubated for 30 min at 37 C. The coating procedure was repeated twice, with fresh solutions used each time. Samples were then incubated with 0.5 M NaOH for 30 min at 37°C and rinsed with MilliQ water five times and allowed to air-dry overnight. Samples were UV sterilised for 20 min on each side prior to cell culture. Scaffolds coated with nnHA were designated T500-H and T1000-H.
