To induce class switching, 200,000 transduced CH12F3-2 cells were seeded into 24-well plate and treated with 1 ng/ml TGF-β1 (Preprotech), 10 ng/ml IL-4 (Preprotech) and 1 μg/ml agonist anti-CD40 (BD). Cells were stained with a PE-conjugated anti-IgA antibody (eBiosciences) and analyzed using a FACSCantoII flow cytometer (BD Biosciences) and Flowing Software.

Quantifying IgA Switching Efficiency

For detecting IgA switching efficiency, 1 3 10 6 cells were stained with 0.1 mg of phycoerythrin (PE)-conjugated anti-IgA antibody (eBioscience, 12-4204-83) and 0.1 mg of APC-conjugated anti-IgM antibody (eBioscience, 17-5790-82) for 5 min at room temperature in 200 mL of 2% FBS/PBS. After washing twice with PBS, cells were adjusted to a concentration of 1-5 3 10 6 cells/mL and were analyzed on a CytoFLEX Flow Cytometer (Beckman Coulter). Flow cytometry data were analyzed using CytExpert software or FlowJo software. Live cells were gated on the FSC/SSC plot and IgA+ cells were further identified on the IgA PE/IgM APC plot.
