Characterization of UT-SCC Cell Lines

The UT-SCC cell lines were provided by Dr. Reidar Grénman (Turku University Hospital, Turku, Finland). All are from a series of HPV-negative cell lines which were developed from primary and recurrent HNSCC specimens during the 1990s [30] , [31] . These cell lines have been maintained at low passage number (<20) and we have used these cell lines extensively in a number of studies where the cell lines, and xenografts obtained from them, have maintained consistent and reproducible characteristics [32] (link), [33] (link), [34] (link), [35] (link), [36] (link), [37] (link), [38] (link), [39] (link), [40] (link), [41] (link), [42] (link), [43] (link). In addition, other researchers have utilized these cell lines [44] (link), [45] (link), [46] (link), [47] (link), [48] (link), [49] (link), [50] (link) and shown similar radiation responses and other characteristics to those reported in our studies. Cells were cultured and maintained in Dulbecco's modified Eagle's medium supplemented with 10% fetal bovine serum, penicillin (100 U/ml), and streptomycin (100 mg/ml) and maintained at low passage number. Dacomitinib and gedatolisib was kindly provided by Pfizer (NY, USA). A 10 mM solution of each was prepared in dimethyl sulfoxide and stored at −70 °C for in vitro experiments.

Preparation of Drug Stock Solutions

Trametinib (GSK1120212) and dabrafenib (GSK2118436) were kindly provided by GlaxoSmithKline (Brentford, Middlesex, UK). Gedatolisib (PF05212384) was kindly provided by Pfizer Inc. (New York, NY, USA). SCH772984 and alpelisib were purchased by Selleckchem (Houston, TX, USA). Everolimus was obtained from Novartis Pharma (Basel, Switzerland). MK-2206 was kindly provided by Merck and Co. (Kenilworth, NJ, USA).
Trametinib, dabrafenib, SCH772984, alpelisib and MK-2206 were dissolved in DMSO as 1 mM, 10 mM, 0.2 mM, 10 mM and 1 mM stock solution, respectively and stored at −20 °C. Gedatolisib was dissolved in DMSO as 1 mM stock solution and stored at −80 °C. Everolimus was dissolved in 100% ethanol as a 10 mM stock solution and stored at −20 °C.
The final concentration of drugs was obtained by dilution with culture medium.

Cell Viability Assay for Drug Screening

Dissociated cells were seeded at 5,000 cells per well to 96-well plates; serially diluted drugs were added and cells were cultured for 3 days. Cell viability was measured by MTS cell viability assay with CellTiter 96 Aqueous One Solution (Promega, Madison, WI) to determine effective concentration 50 (EC50) values. Experiments were performed in triplicate. Targeted agents tested include the EGFR and HER2 inhibitor lapatinib (Tykerb®, GlaxoSmithKline, Brentford, UK), the phosphatidylinositol-3 kinase (PI3K) and mammalian target of rapamycin inhibitor PF-05212384 (gedatolisib) (Pfizer), the mitogen-activated protein kinase (MAPK)/Erk kinase (MEK) inhibitor PD98509 (Cell Signaling), and the mesenchymal-epithelial transition (Met) and anaplastic lymphoma kinase inhibitor crizotinib (Xalkori®, Pfizer, New York, NY).

Gedatolisib Pharmacological Evaluation

Gedatolisib (also known as PKI‐587 or PF‐05212384) was kindly provided by Pfizer (New York, NY). CHIR‐99021 (HCl) and SB216763 were purchased from Selleck Chemicals (Houston, TX). LiCl was purchased from Sigma Aldrich (St Louis, MO). Stock solutions were prepared in dimethyl sulfoxide or distilled water and stored at −20 °C.

Gedatolisib Injection Dosage Study in Nude Mice

8‐ to 9‐week‐old female, Crl:NU(NCr)‐Foxn1nu mice (Charles River, Strain 490, Wilmington, MA, USA) were injected with either vehicle, 1 or 10 mg·kg−1 Gedatolisib (Pfizer, PF‐05212384, New York, NY, USA) via the tail vein. Gedatolisib vehicle consisted of 0.3% l‐lactic acid (Sigma, L6402) and 5% dextrose (Sigma, D9434) in endotoxin‐free ultrapure water (Millipore, TMS‐011‐A, Burlington, MA, USA) and was sterile‐filtered through a 0.2 μm PES filter. Gedatolisib was added to vehicle at 2 mg·mL−1, heated at 60 °C for 15–20 min, and then sterile‐filtered. Sterile PBS was used for balance, for a final injection volume of 100 μL. Animals were euthanized 1 or 2 h following injection.
