Biomarker Measurement in Metabolic Study

Assessing Renal Function: Serum Creatinine and BUN

Renal functions were assessed by measuring serum creatinine and blood urea nitrogen (BUN) levels. Briefly, the blood samples were initially kept at room temperature for about 20 minutes, following which they were centrifuged to collect the clear serum. Serum creatinine and BUN levels were evaluated to determine renal functions with serum creatinine kit (Jiancheng, China, catalog C011-2-1) and BUN kit (Jiancheng, China, catalog C013-2-1). For serum creatinine measurement, standards, serum samples and blank water were alternatively mixed with 180μl Enzyme Solution A in the 96-well plate, which was incubated in a 37°C incubator for five minutes. After that OD546nm readings were retrieved. 60μl Enzyme Solution B was added, and the plate was maintained at 37°C for another 5 minutes. The second OD546nm readings were retrieved. The concentration of serum creatinine levels was calculated accordingly. For serum BUN levels, 20μl serum samples, standards or blank water were alternatively mixed with 250μl urease solution. The mixtures were maintained for 10min at 37°C. Phenol solution (1ml) and sodium hypochlorite solution (1ml) were then added, and the tubes containing the mixtures were immersed in 37°C water bath for 10 minutes. Finally, OD640 readings were retrieved, which were used for the calculation of BUN levels.

MB was prepared by the sonication-lyophilization method as reported in our recent study [46 (link)].

Measuring Urea and Albumin Secretion

Comprehensive Renal Assessment Protocol

The levels of blood urea nitrogen (BUN), serum and urine creatinine (Scr, Cre), urinary protein (UPr), TNFα, IL1β, IL-5 and IL-6 in serum and urine of rats were determined by ELISA. All steps were strict to the instructions. After completing the last step, the absorbance at the specified wavelength was detected using a multifunctional enzyme-labeled analyzer (MB-530, Heales) within 5 min. The serum and urinary creatinine were measured by the creatinine kit (#C011-2-1, Jiancheng) and the Creatinine clearance rate (Ccr= Cre × urine volume per minute/Scr) was calculated accordingly. The BUN kit (#C013-1-1), and UPr kit (#C035-2-1) were purchased from Nanjing Jiancheng Bioengineering Institute. Sino-American Biotechnology Co., Ltd provided the TNFα kit (#CSB-E11987r), IL1β kit (#CSB-E08055r), IL-5 kit (#CSB-E07435r), and IL-6 kit (#CSB-E04640r).
