Cell Cycle Analysis of HUES6 Cells

For cell cycle analysis, HUES6 cells were individualized using accutase 1:4 in PBS and fixed in 70% ethanol overnight. For BrdU detection, cells were incubated with 10 μM BrdU for 30 minutes before collection. After incubation with 0.5 mg/ml pepsin cells were treated with 2 N HCl for 20 minutes, washed and incubated with a rat-antiBrdU antibody (AbD serotec) for 45 minutes followed by incubation with goat anti-rat-FITC secondary antibody (Southern Biotech) for 30 minutes. Cells were then resuspended in PBS containing 50 μg/ml of propidium iodide and 0.5 mg/ml of RNaseA.
