The complete genome was sequenced using 454 technology with the GS Junior 454 system platform (Roche Applied Science, Indianapolis, IN, USA), yielding 6.5-fold coverage of the phage genome [68 (link)]. Prediction of open reading frames (ORFs) was performed using softberry (http://linux1.softberry.com/ (accessed on 8 February 2021)) and Glimmer. A whole-genome sequence assembly was performed using Geneious software version 6 while Phage termini were identified using the method described by Zhang et al. [69 (link)].

Complete Genome Sequencing of Mycobacteriophage

Check if the same lab product or an alternative is used in the 5 most similar protocols
The complete genome was sequenced, using 454 technology, with the GS Junior 454 system platform (Roche Diagnostics, Indianapolis, IN, USA), yielding 145-fold coverage of the phage genome. The 15 available nucleotide sequences of the mycobacteriophage genomes in the F cluster were retrieved from the NCBI nucleotide database. Based on the phage coding sequence, a phylogenetic tree was drawn, using the neighbor linkage (NJ) method. The bootstrap value was 1000 replicates on MEGA X [19 (link)].

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
