Acclimation of Male Sprague-Dawley Rats

Male Sprague-Dawley rats (N = 10) were obtained from Envigo at 7 to 8 weeks of age. TEK-Fresh® (Envigo, Indianapolis, IN) bedding was used in polycarbonate home cages (48.3 × 26.7 × 20.3 cm [l × w × h]) where each rat was housed individually. Upon arrival, rats were handled daily for one week to acclimate them to the colony and personnel. Before the start of the study, food was restricted to maintain weights at 85% free-feed body weight (average ad lib feed weight = 291 g). This weight was increased by 2 g each month. The colony room was temperature- and humidity-controlled with lights on a 12-hour light/dark schedule (lights on at 6 AM). All protocols were approved by the University of Nebraska-Lincoln Institutional Animal Care and Use Committee.

Sprague Dawley Rats for Experimental Research

Housing and Feeding of Sprague-Dawley Rats

Circadian Rhythm Study in Rats

Influences of Age, Housing, and Context on Rats

Male Sprague Dawley rats (Envigo Inc, Haslett, MI) arrived at post-natal day 25, P25 (60–80 gm, n=51), or P56 (260–280 gm, n=32) and were pair housed upon arrival with ad libitum access to food and water. Humidity and temperature were regulated within the vivarium and rats were maintained on reverse lighting conditions (7 am off, 7 pm on) with all behavioral conditions occurring during the rats’ active cycle. Rats underwent habituation and handling for 7–8 days prior to surgery. Following surgical procedures, rodents were separated into two housing groups (pair vs single) and maintained on 15 gm (adolescent) or 18 gm (adult) of chow, which were adjusted to allow for normal growth (Carroll ME 1985 (link); DePoy et al. 2016 (link); Cho et al. 2018 (link)). Female rats were not included in the current study to focus on Age x Context × Housing factors. All protocols were approved by the Institutional Animal Care and Use Committee (IACUC) at Michigan State University (MSU) and followed the National Research Council’s Guide for the Care and Use of Laboratory Rats.

Traumatic Brain Injury in Juvenile Rats

Male Sprague Dawley rats (Envigo, Indianapolis, IN) were used for all experiments. Rats were housed in a 12 h light: 12 h dark cycle at a constant temperature (23°C ± 2°C) with food and water available ad libitum according to the Association for Assessment and Accreditation of Laboratory Animal Care International guidelines. All rats were acclimated from shipping a minimum of one week prior to experiments. PND17 rats were shipped with the dam. After surgery, post-operative care via physical examination took place to monitor each animal's condition. PND17 rats were returned to their dam following surgery and midline fluid percussion injury (mFPI) until tissue collection (2H, 24H, 7D), or until they were weaned at PND24. Average PND17 pre-surgical weight was 28.2 ± 3.2 g. Average PND35 pre-surgical weight was 135.5 ± 14.3 g. Weights and health conditions were monitored and documented throughout the experiment. Animal care and experiments were approved by the Institutional Animal Care and Use Committee (IACUC) at the University of Arizona (protocol 13–460).
