SIV Infection in Rhesus Macaques

Purpose-bred adult rhesus macaques (Macaca mulatta) of Chinese origin were housed at the German Primate Center (DPZ) according to the German Animal Welfare Act which complies with the European Union guidelines on the use of NHPs for biomedical research and the Weatherall report. The study was approved by the Lower Saxony State Office for Consumer Protection and Food Safety and performed with the project license 509.42502/08-04.03. Six animals were infected intravenously (i.v.) with cell-free SIVmac251. Three of those animals were additionally inoculated i.v. with cell-associated SIVmac251, as described elsewhere26 (link). Transmission of cell-associated virus in these animals accelerated the initial virus dissemination26 (link). Colonic biopsies were collected seven days before and at several time points after infection under combined ketamine, xylazine and atropine anaesthesia as previously described26 (link),42 (link). One day before colonoscopy, animals received Bisacodyl orally in water (1.2 – 2.7 mg/kg; Dulcolax, Sanofi-Aventis Germany) to stimulate gut motility. Bowel Preparation Protocol for Capsule Endoscopy

Bowel preparation for CCE was a split dose 4-L (4L) Polyethylene glycol (PEG) (KleanPrep—Norgine, Middlesex, UK) with the addition of Phospho-soda, “Fleet” (Casen Recordati, Zaragoza, Spain), Gastrograffin (Bayer, Reading, UK) and rectal Bisacodyl (Dulcolax—Sanofi-Aventis, Ireland) suppository booster medications. Similarly, all patients received a further split dose 4L Polyethylene glycol (KleanPrep—Norgine, Middlesex, UK), bowel preparation prior to colonoscopy (Appendix C). In addition, standard dietary manipulation was advised for several days prior to both procedures.
All obtained data were kept on secure encrypted hospital computers. Statistical analysis was performed using MedCalc (MedCalc Software Ltd, Belgium). A Chi-squared test was used to compare categorical data and a Student t-test was used to compare means, a p < 0.05 was deemed significant. Pearson Correlation Coefficient was used to assess the correlation between patient and operator reported comfort scores. A r value of ≥ 0.7 was considered as a strong correlation.

Colon Cleansing with Colon Hydromat

The Colon Hydromat (manufacturer Herrmann Apparatebau GmbH, Germany), is a CE marked device marketed in Europe that is intended for colon cleansing when medically indicated, such as before colonoscopy. Colonic irrigation with the Colon Hydromat uses constant warm water lavage with a contained temperature and pressure control. The device cleanses the large bowel through a series of fill and empty cycles by hydrating the colon, stimulating peristalsis, and evacuating bowel contents without the use of further chemicals. In addition, by using a complete closed system, the method is discrete and without odor or embarrassment for the patient.

Colon Preparation for AHDS Dogs

A nasoesophageal tube was placed in all AHDS dogs and secured. All colon preparation medications were administered via the nasoesophageal tube in all AHDS dogs. At time 0, 20 mg bisacodyl (Dulcolax®, Sanofi, Bridgewater, NJ) mixed in 4 mL/kg lukewarm water was administered. At times 1-hour and 5-hour, 1 packet of polyethylene glycol (Klean Prep®, Helsinn-Birex Pharmaceuticals Ltd, County Dublin, Ireland) dissolved in 4 mL/kg of lukewarm water was administered. For times 2–4 and 6-8-hours, 4 mL/kg lukewarm water was administered every hour.
