Piezo Mechanosensitive Ion Channels in Mice

All animal experiments were carried out according to protocols approved by the Harvard Medical School Institutional Animal Care and Use Committee. Mice described in the literature and purchased from the Jackson Laboratories: Piezo1P1-tdT (stock# 029214) (Ranade et al., 2014 (link)), Piezo2-EGFP-IRES-Cre (Piezo2tm1.1(cre)Apat, stock# 027719) (Woo et al., 2014 (link)), Piezo1f/f (stock# 029213) (Cahalan et al., 2015 (link)), Piezo2f/f (stock# 027720) (Woo et al., 2014 (link)), Prrx1-Cre (stock# 005584) (Logan et al., 2002 (link)), Sp7-GFP::Cre (stock# 006361) (Rodda and McMahon, 2006 (link)), Polr2aTn(pb-CAG-GCaMP5g,-tdTomato)Tvrd (PC::G5-tdT, stock# 024477) (Gee et al., 2014 (link)), Ai9 (Rosa26-TdTomato, stock# 007909). For embryos or neonatal mice, both male and female were used in the analyses as sex could not be clearly identified in embryos or neonatal mice. Sex-matched littermate mice were compared in postnatal mice at day 21. Three or more littermate groups were examined and representative images are shown.

Mouse Genetic Models for Neuroscience Research

All animal procedures followed the ethical guidelines outlined in the National Institutes of Health Guide for the Care and Use of Laboratory Animals, and all protocols were approved by the institutional animal care and use committee (IACUC) at Harvard Medical School. Mice were maintained under constant temperature (23 ± 1 °C) and relative humidity (46 ± 5%) with a 12-h light:dark cycle. Gabra1-IRES-cre, lsl-L10-GFP and Ptger3flox mice were previously generated14 (link),22 (link),51 (link).

Lineage Tracing of Sox10-Expressing Cells

All of the mice used in this study were housed in a strict pathogen‐free environment. Sox10‐Cre (#025807), Sox‐CreERT2 (#027651), Piezo2‐EGFP‐IRES‐Cre (#027719), and Ai9 (#007909) mice were originally purchased from the Jackson Laboratory (Bar Harbor, ME, USA). Rosa26‐mTmG (mT/mG) and wild‐type C57BL/6J mice were acquired from Prof. Ren Xu (Xiamen University). CTSK‐Cre mice were obtained as described in previous research.[107] All of the mice analyzed were maintained on the C57BL/6 background. For lineage tracing, Sox‐CreERT2;mT/mG mice were generated and intraperitoneally injected with tamoxifen (20 mg mL−1, T5648; Sigma‐Aldrich, St. Louis, MO) in corn oil (#C8267, Sigma‐Aldrich) daily for 1 and 3 d at the indicated time points. All of the animal experiments were conducted according to guidelines approved by the institutional animal care and use committee at Xiamen University (XMULAC20190084).

Cre-Dependent Mouse Lines for Neuroscience Research

Crhr2-ires-Cre and Gabra1-ires-Cre mice were constructed as described below; Gpr65-ires-Cre, Glp1r-ires-Cre, Npy2r-ires-Cre, P2ry1-ires-Cre and P2×2/P2×3−/− mice were described before (Chang et al., 2015 (link); Finger et al., 2005 ; Williams et al., 2016 (link)); Vglut2-ires-Cre and loxP-L10-GFP mice were gifts from Bradford Lowell (Beth Israel Deaconess Medical Center); and wild type C57BL/6J (000664), Calb1-ires-Cre (028532), Chat-ires-Cre (006410), Chat-GFP (007902), Krt8-CreER (017947), Npy1r-Cre (030544), Piezo2-EGfp-ires-Cre (027719), Plcβ2−/− (018064), loxP-tdTomato (007908), loxP-ChR2 (012569), and loxP-DTR (007900) mice were purchased (Jackson Laboratory). The constitutive GCaMP3 allele (Rosa26-GCaMP3) was generated by breeding loxP-GCaMP3 (014538) with E2a-Cre mice (Jackson, 003314), and then crossing out the E2a-Cre allele. Single-cell sequencing was performed on male C57BL/6J mice; both male and female mice between 8–24 weeks old were used for all other studies, and no differences based on sex were observed.

Genetic Mouse Models for Mechanosensation

All experiments involving mice were approved by the Institutional Animal Care and Use Committee (IACUC) of the Weizmann Institute. Mice were housed in a temperature- and humidity-controlled vivarium on a 12 hr light-dark cycle with free access to food and water.
The following strains were used: Piezo2EGFP-IRES-Cre (The Jackson Laboratory, #027719), Gli1CreERT2 (The Jackson Laboratory, #007913), Rosa26tdTomato (The Jackson Laboratory, #007909), and Thy1-YFP16 (The Jackson Laboratory, #003709).
In all experiments, at least three mice from different litters were used. Mice were genotyped by PCR of genomic DNA from ear clips. Primer sequences and amplicon sizes are listed in Table 1.
