Immunohistochemical Analysis of Tumor Tissues

Tissues were fixed in formalin and embedded in paraffin, and the sections were used for hematoxylin-eosin staining and IHC. For IHC, the sections were subjected to heat-mediated antigen retrieval in pH 6.0 citrate buffer after routine deparaffinization and rehydration. The following staining steps were conducted using a rabbit biotin-streptavidin horseradish peroxidase detection system (SP-9001, ZSGB-BIO, China). The staining images of tumor tissues were evaluated by two researchers, according to the staining intensity. The CD31 antibody (#77699, CST, USA) and endoglin antibody (ab221675, Abcam, UK) were diluted at 1:100. The ADAMTS2 antibody (PA5-78728, Invitrogen, USA) was diluted at 1:500. Immunohistochemical Analysis of FTO

IHC was performed as previous described27 and was employed to determine the expression of FTO protein in PC clinical specimens. The tissues were fixed with 4% paraformaldehyde, embedded in paraffin, and sectioned at 3 μm. Then use rabbit polyclonal anti‐THBS2 antibody (1:250, abclonal, A8561, China), ADAMTS2 Antibody (1:25, Invitrogen, PA5‐50539, USA), COL12A1 (1:50, abcam, ab121304, UK), and anti‐FTO antibody (1:250, abclonal, A3861, China) at 4°C overnight. After the secondary antibody was incubated, the sections were stained with the DAB reagent. The results were analyzed using Image‐Pro Plus 6.0 software.
