Isolation and Differentiation of Mouse Brown Adipose Tissue Stromal Vascular Fraction

Primary BAT SVF cells were isolated from C57BL/6J background mice as described (Shan et al., 2016 (link)). Briefly, the interscapular brown fat was dissected from 8-week-old mice. Next, the brown fat was cut to fine pieces and digested with 1.5 mg/ml collagenase type I (catalog number SCR103, Sigma-Aldrich) at 37°C water bath for 50 min. The tissue was vortexed every 10 min. Then, the digestion was filtered through 100-μm and 70-μm cell strainers. The digestion was centrifuged at 400 g for 6 min to enrich SVF cells. The cells were seeded into a cell culture plate with growth medium (DMEM and 10% fetal bovine serum). To induce adipogenic differentiation, the growth medium was supplemented with 2.85 mM recombinant human insulin (catalog number I8830, Solarbio), 0.3 mM dexamethasone (catalog number D8040, Solarbio), and 0.63 mM 3-isobutyl-methylxanthine (catalog number I7018, Sigma-Aldrich). After 96 h, the medium was switched to growth medium, supplemented with 10 nM triiodothyronine (catalog number T6397, Sigma-Aldrich), and 200 nM insulin to induce mature adipocytes. For Oil red O staining, cells were fixed with 10% formaldehyde for 5 min and stained with staining solutions (catalog number G1262, Solarbio) according to the instructions.

Isophane Protamine Biosynthetic Human Insulin Injection (Pre-mixed 30R) was purchased from Novo Nordisk (China) Pharmaceuticals Co., Ltd.

Glucose Homeostasis Assessment in Rats

For OGTT, rats were fasted for 12 h after which blood glucose was measured using the tail clipping method, and followed by gavage with distilled water or 50% glucose at 2 g/kg body weight. Blood glucose levels were measured at indicated time point. For ITT, rats were fasted for 6 h during the light cycle. Distilled water or insulin (INS) (recombinant human insulin, Solarbio, Beijing, China, 0.75 units/kg) was injected intraperitoneally, and blood glu-cose levels were assayed at indicated time points after insulin injection. Blood glucose levels were measured using Roche's Blood Glucose Meter and Test Strips (AUUC-CHEK Performa Zhuoyuejingcai Glucose Meter, Roche Diabetes Care GmbH, Mannheim, Germany).

Glucose Consumption Assay in HepG2 Cells

HepG2 cells were plated at a concentration of 2 × 104 per well in 96-well plates and incubated for 24 h at 37 °C. Afterward, cells were treated with PA/BSA complex with or without LPMP for 24 h. Then, HepG2 cells were further cultured for 4 h in phenol red-free MEM medium with or without 200 nm human recombinant insulin (Beijing Solarbio Science & Technology Co., Ltd., Beijing, China). Subsequently, the medium was collected and the glucose residue was determined by the glucose oxidase-peroxidase method according to the Trinder reaction [67 (link),68 (link)]. The glucose consumption was obtained by subtracting the remaining glucose content from the total glucose content in the culture medium.
