Synthesis of PEG-Polylysine Conjugates

PEG-PLys, used for constructing PM as described in the following section, was synthesized via ROP of l-Lys(TFA)-NCA using PEG-NH2 (Mn = 12 kDa) (NOF Corporation) as a macroinitiator. The Mw/Mn of PEG-PLys(TFA) was 1.05 according to SEC. The DP of PLys in PEG-PLys was 44, based on the 1H NMR spectrum. The 1-imino-4-mercaptobutyl (IM) groups were introduced onto the side-chain ε-amino groups of the lysine units of the PLys segment in PEG-PLys [PEG-PLys(IM)] using 2-iminothiolane (Thermo Fischer Scientific), according to a previous report (39 (link)). The introduction ratio of IM in the total NH2 groups in the original PEG-PLys was 50%, according to the 1H NMR.
