A Milli-Q gradient water system was used to produce ultra-pure water. Normal human lung fibroblast cells (MRC-5 cells) and adenocarcinoma human alveolar epithelial cells (A549 cells) were obtained from the American Type Culture Collection (ATCC). All materials detailed herein were used without further purification.

Apoptosis Quantification via Annexin V-FITC/PI

After treating cells with OSW-1 for 24 h, the cells were digested to generate single-cell suspensions, which were stained according to the kit instructions. The experiment was replicated three times for each measurement.

Melatonin Modulates Apoptosis and PD-L1 in NSCLC

To detect cell apoptosis, NSCLC cells (5 × 105 cells/well) were seeded into 6-well plates and treated with melatonin for 24 h. Cells were trypsinized followed by incubation with an Annexin V-FITC/propidium iodide (PI) Apoptosis detection kit (Elabscience) according to the manufacturer’s instruction. To measure PD-L1 protein levels, cells were treated with melatonin (2.5 mM) in the presence or absence of IFNγ (50 ng/mL) for 24 h. Cells were then trypsinized followed by incubation with a PE-conjugated anti-PD-L1 antibody (Abcam) and analyzed with a BD FACSVia flow cytometer (BD Biosciences).
