Adipocyte Differentiation Assay Protocol

Exosome-depleted FBS and exosome marker antibodies including anti-CD9, anti-CD81, anti-CD63, and anti-Hsp70 were procured from System Biosciences (Palo Alto, CA, USA). Anti-LAMP1 antibody was purchased from Abcam (Cambridge, MA, USA).

Extracellular Vesicle Characterization in Prostate Cancer Cell Lines

Secondary antibodies anti-rabbit IgG (H + L) F(ab’)2 fragment (Alexa Fluor® 555 Conjugate) (#4413 S), anti-mouse IgG (H + L) F(ab’)2 fragment (Alexa Fluor® 647 Conjugate) (#4410 S), and anti-mouse IgG (H + L) F(ab’)2 fragment (Alexa Fluor® 488 Conjugate) (#4408 S) were from Cell Signaling Technology (Danvers, MA). RPMI1640, penicillin and streptomycin, 0.25% trypsin, fetal bovine serum (FBS), and heat-inactivated FBS were from Gibco Laboratories (Gaithersburg, MD). Halt™ Protease and Phosphatase Inhibitor Cocktail, Keratinocyte-SFM and BRFF-HPC1™ media were from Thermo Fisher Scientific (Waltham, MA). DC™ Protein Assay kit was from Bio-Rad (Hercules, CA). The Poly- L-Lysine, Lactate Assay Kit II, 5-(N,N-Dimethyl)amiloride hydrochloride (DMA) and GW4869 were from Sigma-Aldrich (St. Louis, MO).

In vitro Screening of FDA-Approved Drugs
