Glucose and Insulin Tolerance Testing

Animals fasted for 4–5 hours and received Myrcludex B or placebo injection 3 hours prior to the experiment. Mice received glucose (2 g/kg) by oral gavage or insulin (1.2 mU/kg) by intraperitoneal injection. Blood was drawn by the tail vein at 0, 15, 30, 60, and 90 minutes, and for the oral GTT also at 120 and 180 minutes. Glucose levels were determined in whole blood using a Contour XT glucometer (Bayer BV, Mijdrecht, the Netherlands). Blood collection for the insulin measurements occurred during the oral GTT.
