The 3,4′,5-trihydroxy-trans-stilbene (resveratrol; purity ≥ 99%), L-glutamic acid, Dulbecco’s modified Eagle’s medium (DMEM), and fetal bovine serum (FBS) were purchased form Sigma-Aldrich (St. Louis, MO, USA). The 3-(4,5-dimetylthiazol-2-yl)- 2,5-diphenyltetra-zoliumbromide (MTT) was purchased from Bio Basic (Markham, ON, Canada). The 2′,7′-dichlorodihydrofluorescein diacetate (H2DCFDA) was obtained from Molecular Probes (Eugene, OR, USA). The Trizol reagent was purchased from Invitrogen (Carlsbad, CA, USA). Modulation of MAPK Signaling by Resveratrol

(St. Louis, MO, USA). U0126, SP600125, and SB203580 from Selleck. cn (Shanghai, China) were dissolved in 0.1% DMSO, which was used as a solvent control. Primary antibodies including Sirt1 and mitogen-activated protein kinases (MAPKs) proteins such as p-ERK, JNK, and p38 MAPK were obtained from CST (Shanghai, China), t-ERK/JNK/p38 MAPK from Proteintech (Wuhan, China), GFAP from Millipore (MA, USA), and GAPDH from Abcam (Shanghai, China). Immunoblot Analysis of Lipid Metabolism Markers

Resveratrol (3, 4′, 5-trihydroxy-trans-stilbene) and 3-methyladenine (3-MA; PI3K class III inhibitor) were procured from Sigma-Aldrich (St. Louis, MO, USA). Recombinant human TNF-α (R&D Systems, Inc., Minneapolis, MN, USA) was used to stimulate the Ca9-22 cells. For in situ hybridization, probes against human SREBP1 (ACD# 469871), human p62/SQSTM1 (ACD# 415881), and human E-FABP (ACD# 566111-C3) were procured from Advanced Cell Diagnostics Inc. (ACD; Newark, CA, USA).

Guinea Pig Dietary Metabolite Supplementation

Guinea pigs (Dunkin-Hartley), weighing 400-800 g, were fed ad libitum on standard Guinea pig/rabbit pellets, with fresh cabbage leaves and lucerne hay roughage. Animals were maintained on a 12 h light/dark cycle. All experiments were conducted in accordance with the guidelines of the National Health and Medical Research Council of Australia and were approved by the institutional (UNSW) Animal Care Ethics Committee.

CACA (cis-4-aminocrotonic acid) and resveratrol (3,4′,5trihydroxy-trans-stilbene) were purchased from Sigma Aldrich (St Louis, MO) and TPMPA (1,5,6-tetrahydropyridin-4-yl)methylphosphinic acid) were purchased from Tocris Cookson, (UK). (±)-cis-(3-Aminocyclopentyl) (butyl)phosphinic acid (cis-ACPBPA) was synthesised as published previously (Hanrahan et al., 2006) , as was cis-2-(aminomethyl)-1carboxycyclopropane (±)-CAMP (Duke et al., 1998) and S-and R-4-amino-1-cyclopent-1-en-1-yl(butyl)phosphinic acids, (S-and R-4-ACPBPA) (Kumar et al., 2008) . All other reagents were of AR grade.
