Comprehensive Immunophenotyping of T-cells and DCs

T-cell suspensions were isolated from the spleen and lymph nodes, and were stained with antibodies against the following cell surface antigens: APC-conjugated anti-mouse CD3, FITC-conjugated anti-mouse CD4, PE-conjugated anti-mouse CD8, PE-conjugated anti-mouse Gr-1, APC-conjugated anti-mouse CD11b, PE-conjugated anti-mouse TCR (eBioscience), PE-conjugated anti-mouse PD-1, and PE-conjugated anti-mouse CD152 (CTLA-4; BD Bioscience, San Jose, CA, USA). Tregs were detected from the spleen and lymph nodes using a Mouse Regulatory T-Cell Staining Kit (eBioscience) according to the manufacturer's instructions. DCs were isolated from the bone marrow and cells were incubated with FITC-conjugated anti-CD86, APC-conjugated anti-CD11C, PE-conjugated anti-MHC-II, FITC-dextran, PE-conjugated anti-CCR7, and anti-PD-L1 (eBioscience). The cells were analyzed by FACS, and the acquired data was performed with FlowJo Software, version 9.1 (Tree Star, San Carlos, CA, USA).

Characterization of Adipose-Derived Immune Cells

Epididymal fat pads were isolated after saline perfusion, then minced, washed, and centrifuged (1,000 g for 5 min). Floating adipose tissue was collected and digested with type I collagenase solution (1 mg/ml, 30 min). This digest was pelleted to yield the SVF, which was washed twice in FACS wash buffer (PBS supplemented with 4% FBS), then blocked for Fc receptors using anti-mouse CD16/32 (BD Bioscience), followed by staining with APC-conjugated anti-mouse CD11b, eFluor 450- conjugated anti-mouse CD11c, and PE-conjugated anti-mouse F4/80 (eBioscience). Other surface markers included CD18 and ICAM-1 (both PE-conjugated, eBioscience). Samples were analyzed using a BD LSD II flow cytometer.

Competitive Repopulation Assay of Phlda3 Genotypes

Phlda3+/+ and Phlda3−/− littermates (CD45.2) 35 days after 2.5 Gy TBI were used as donors for the competitive repopulation assay. 2 × 105 or 1 × 106 whole bone marrow cells from these mice were mixed with 2 × 105 whole bone marrow cells from unirradiated B6.SJL (CD45.1) mice. The mixed bone marrow cells were transplanted into 8-week-old B6.SJL (CD45.1) recipients 6 h after two fractions of 4.75 Gy TBI with an interval of about 18 h. At 16 weeks after bone marrow transplantation, peripheral blood mononuclear cells (PB-MNCs) were harvested from mice following the procedure described previously3. PB-MNCs were blocked with a rat anti-mouse CD16/32 antibody (BD Pharmingen) and stained with an antibody cocktail including PE-Cy5 conjugated anti-mouse CD3e (clone: 145-2C11), PE conjugated anti-mouse B220 (clone: RA3-6B2), APC conjugated anti-mouse CD11b (clone: M1/70), FITC conjugated anti-mouse CD45.2 (clone: 104), and APC-eFluor780 conjugated anti-mouse CD45.1 (clone: A20) antibodies (eBioscience). All antibodies were diluted 1:400. Data were collected from at least 20,000 single cells by FACSCanto (BD Pharmingen) and analyzed by FlowJo (Tree Star, Inc) without knowledge of the genotype or treatment by a single observer (C-LL).
