In Vitro Glucuronidation of Emodin and its Derivatives

Bilirubin (99.3%), Trizma base, and alamethicin were obtained from J&K Scientific (Beijing, China). Uridine diphosphoglucuronic acid (UDPGA), d-saccharic acid 1,4-lactone, and MgCl2 were purchased from Sigma-Aldrich (Beijing, China). Pooled male Sprague–Dawley RLM (452501) were obtained from BD Gentest (Shanghai, China). Emodin (99.3%), physcion (99.2%), and rhein (99.8%) were purchased from the National Institute for Food and Drug Control (Beijing, China). Emodin-8-O-glc (98.0%), citreorosein (98.0%), trans-emodin dianthrones (98.0%), and cis-emodin dianthrones (98.0%) were purchased from Shanghai Yuanye Bio-Technology Co. (Shanghai, China). Polygonumnolides C2, C3, and C4 and the P. multiflorum extract were gifts from Dr. Yang (Institute of Materia Medica, Chinese Academy of Medical Sciences & Peking Union Medical College). Polygonumnolides C2, C3, and C4 were isolated and characterized by Dr. Yang and colleagues20 (link). All other chemicals used in the glucuronidation incubations and the ultra-performance liquid chromatography (UPLC) solvents were of high-performance LC grade and were obtained from Sigma-Aldrich (Beijing, China).

(Beijing, China). Human liver microsomes (HLMs) were acquired from Rild Liver Disease Research Co., Ltd (Shanghai). All analytical solvents were of liquid chromatography (LC) - grade and were obtained from Sigma-Aldrich (St. Louis, USA) or Merck (Darmstadt, Germany).

In vitro UDP-glucuronosyltransferase Assay

Synthesis and Characterization of Key Biochemical Reagents

The nucleotide monophosphate RMP (purity of 93%) and nucleotide triphosphate RTP (purity of 98%) were synthesized as previously reported [13 (link)]. Dimethyl sulfoxide (DMSO), 5,5′-dithiobis (2-nitrobenzoic acid) (DTNB, ≥ 98%), Trizma® base (Tris, ≥99.9%) and magnesium chloride hexahydrate (MgCl2·6H2O, 99%–102%) were purchased from Sigma-Aldrich (St. Louis, MO, USA). PhosSTOP EASYpack (PhosSTOP) was purchased from Roche (Basel, Switzerland). Alamethicin was purchased from J&K Chemical, Ltd. (Beijing, China). Potassium chloride (KCl, ≥99.5%), ethylenediaminetetraacetic acid disodium salt dihydrate (Na2-EDTA·2H2O, ≥99.0%) and sucrose were obtained from Sinopharm Chemical Reagent Co., Ltd. (Shanghai, China). All other reagents were of the highest quality commercially available.

Schisantherin A Metabolism in Rat Liver Microsomes

Schisantherin A was purchased from the National Institute for Food and Drug Control (Beijing, China), and the purity was over 98% according to the HPLC analysis. Acetonitrile, ammonium acetate, and acetic acid were used for chromatography (HPLC grade; Fisher, Fair Lawn, NJ, USA). Water was purified by a Milli-Q academic purification system (Millipore, ELIX100, Bedford, MA, USA). Analytical grade carboxymethyl cellulose sodium (CMC-Na) was purchased from Kermel Chemical Reagents Development Centre (Kermel, Tianjin, China). β- NADPH(β-nicotinamide adenine dinucleotide 20-phosphate), glucose-6-phosphate, glucose-6-phosphatedehydrogenase, and UDPGA (uridine-50-diphosphoglucuronic acid trisodium salt) were acquired from Sigma Chemical Co. (St. Louis, MO, USA). Alamethicin was purchased from J&K Scientific (Beijing, China). Tris-HCl was obtained from Phygene Life Sciences Co. Ltd. (Fuzhou, China). MgCl2 was from Tianjin Hongyan Chemical Reagent Factory (Tianjin, China). RLMs were laboratory-made at the Department of Pharmaceutical Analysis in the School of Pharmacy, Hebei Medical University, and stored at −80 °C until used.
