Cytokine Signaling Pathway Analysis in Cell Lines

All the reagents were purchased from Sigma-Aldrich (St. Louis, MO, USA). De Man, Rogosa, and Sharpe (MRS) broth was purchased from BD Biosciences (Franklin Lakes, NJ, USA). RAW 264.7, RBL-2H3, and HT-29 cell lines were obtained from the Korean Cell Line Bank (Seoul, Korea). Dulbecco’s modified Eagle’s medium (DMEM), fetal bovine serum (FBS), penicillin–streptomycin (P/S), and phosphate-buffered saline (PBS) were purchased from Gibco (Gaithersburg, MD, USA). HiGene™ Total RNA Prep Kit (BIOFACT, Daejeon, Korea) was reverse-transcribed to cDNA using the SuperScript™ IV First-Strand Synthesis System (Invitrogen, Waltham, MA USA), and FastStart Essential DNA Green Master (Roche, Basel, Switzerland) was used for investigating mRNA expression. RIPA buffer was obtained from iNtRON (Seongnam-si, Gyeonggi-do, Korea) and contained phosphatase and protease inhibitor cocktails (GenDEPOT, Katy, TX, USA). The Pierce BCA Protein Assay Kit was purchased from Thermo Fisher Scientific (Waltham, MA, USA). Antibodies against STAT6, and phosphorylated STAT6 (p-STAT6) were purchased from Cell Signaling Technology (Danvers, MA, USA). Amersham™ Imager 600 (GE Healthcare, Chicago, IL, USA) and WesternBright ECL kit (Advansta, San Jose, CA USA) were used for protein image analysis.

Culturing Rat Basophilic Leukemia and B16 Melanoma Cells

Rat basophilic leukemia (RBL2H3) cells were obtained from the Korea Cell Line Bank (Seoul, Korea). Cells were grown in Dulbecco's modified Eagle's medium containing heat-inactivated fetal bovine serum, 2 mM L-glutamine, 100 units/ml penicillin, and 100 μg/ml streptomycin (Invitrogen). Cultures were maintained in 5% CO2 at 37°C. The isolation of macrophages from lung tissues was done according to the standard procedures [20 (link)]. B16F1 and B16F10 cancer cells used in this study were cultured in Dulbecco's modified minimal essential medium (DMEM; Invitrogen) supplemented with heat-inactivated 10% fetal bovine serum (FBS, Invitrogen) and antibiotics at 37°C in a humidified incubator with a mixture of 95% air and 5% CO2.

Culturing of Colorectal and Mast Cell Lines

Rat Basophilic Leukemia Cell Culture

The rat basophilic leukemia cell line RBL-2H3 was supplied from the Korean Cell Line Bank (Seoul, South Korea). RBL-2H3 cells were cultured in Minimum Essential Medium (MEM) (Welgene, South Korea) supplemented with 10% FBS (Gibco, Rockville, MD, United States) and 1% penicillin-streptomycin under an atmosphere of 5% CO2 in a humidified 37°C incubator. RAW264.7 macrophage cells, IgEL b4 cells and 293 T cells were purchased from American Type Culture Collection (Manassas, VA, United States) and were cultured in Dulbecco Modified Eagle’s medium (DMEM) with 10% FBS and 1% penicillin-streptomycin. Calcium ionophore A23187 and phorbol 12-myristate 13-acetate (PMA) were purchased from Sigma-Aldrich (St. Louis, MO, United States). Quercetin, 1-Chloro-2,4-dinitrobenzene (DNCB) and dexamethasone were purchased from Sigma-Aldrich (St. Louis, MO, United States). Albumin from bovine serum, 2,4-dinitrophenylated (DNP-BSA) was purchased from Invitrogen by Thermo Fisher Scientific (Eugene, OR, United States). rIL-4 and mTNF-α were purchased from R&D system Inc (Minneapolis, MN, United States). mIL-6 was purchased from Biolegend (San Diego, CA, United States).
Stock solutions of ZCO were diluted using the mixture solution of DMSO and ethanol (1:1) to a concentration of 20% and filter-sterilized, and then diluted with phosphate buffered saline (PBS) for the working concentration.

Culturing Mouse Macrophages and Rat Mast Cells

Mast Cell Activation Assay Protocol

RBL-2H3, rat peritoneal mast cells (RPMC), and mouse bone marrow-derived mast cells (mBMMC) were grown at 37°C in 5% CO2 in Dulbecco's modified Eagle's medium, α-minimum essential medium (Gibco, Grand Island, NY) and RPMI 1640 (Hyclone, Logan, UT), respectively, supplemented with 10% heat-inactivated fetal bovine serum, 100 U/mL penicillin G, 100 μg/mL streptomycin, and 250 ng/mL amphotericin B. In addition, complete RPMI 1640 media was supplemented by 4 mM L-glutamine, 25 M HEPES, 50 μM 2-mercaptoethanol, 1 mM sodium pyruvate, MEM non-essential amino acid solution (Gibco), 10 ng/mL murine IL-3, and 2 ng/mL murine SCF (PeproTech, Rocky Hill, NJ). RBL-2H3 were used throughout the study at passages ranging from 4 to 8.
