Visualizing eNOS Localization in ECV304 Cells

Chick Vascular Bed Hypoxia and Ischemia

The chick vascular beds were treated with global hypoxia and ischemic for 2 h. Next, the isolated vascular beds from egg yolk were washed with 1X PBS. The vascular beds were fixed with 4% of paraformaldehyde and permeabilized with 0.1% of Triton X. Finally, they were stained with Alexa Fluor-phalloidin (10 µM) and incubated for 10 min. The vascular beds were then washed with 1X PBS. Images were captured at 20X magnification using Olympus IX71 epifluorescence microscopy system equipped with a DP71 camera.
