Ovulation Synchronization for First TAI

Positive pregnancy diagnosis was made when an amniotic vesicle with viable embryo was detected via rectal palpation with ultrasonography (Honda HS 101V with a 5.0-MHz linear probe, Honda, Tokyo, Japan) 32 and 60 d after TAI.

Estrus Synchronization in Angus Heifers

Virgin Angus heifers across two different years (year 1, n = 38; year 2, n = 28; total n = 66) during the month of April and located at a University of Tennessee AgResearch and Education Center were grazed on mixed grass pastures and had ad libitum access to hay. Average heifer age was 1.2 ± 0.1 yr. Estrus was synchronized according to Figure 1A and defined as the time when a heifer first stood to be mounted by another. Eleven days later, gonadotropin-releasing hormone (GnRH) was administered (100 μg i.m., gonadorelin hydrochloride, Factrel, Zoetis or 100 μg i.m., gonadorelin diacetate tetrahydrate, Cystorelin, Boehringer Ingelheim, Duluth, GA, USA). A controlled internal drug release (CIDR) device was placed intravaginally (1.38 g progesterone, Eazi-Breed CIDR, Zoetis). Seven days later, the CIDR was removed, PGF was administered, and an iButton that had been previously affixed to a blank CIDR (containing no progesterone) was placed intravaginally. Beginning ~24 h after PGF, estrus activity was visually assessed hourly until the first heifer displayed signs of estrus at which point monitoring was continuous by a team of individuals on a rotating schedule.

Breeding Protocols for TAI + NM Cows

The TAI + NM cows were synchronized using a protocol based on the use of a device with 1.9 g of natural progesterone (CIDR 1900 Cattle Insert, Zoetis, Mexico) and the administration of 2 mg IM of Estradiol Benzoate (Estradiol Benzoate, Zoetis, Mexico) on the day of insertion (60-day postpartum). At CIDR withdrawal (67-day postpartum), 400 UI IM of ECG were given (Novormon 5000, Zoetis, Mexico) together with 25 mg IM dinoprost tromethamine (Lutalyse, Zoetis, Mexico). This treatment was followed by 1 mg Estradiol Benzoate at day 68 postpartum. Thus, artificial insemination with the same commercial Brahman bull was performed at 56 ± 2 h after CIDR withdrawal. Then, 20 days after timed artificial insemination the cows were relocated to a natural mating program with a different bull.
The NM cows remained with a 5-year Brahman bull with proven fertility and acceptable following the breeding soundness evaluation in a paddock from day 60 ± 5 to 120 postpartum.

Double-Ovsynch Protocol for Estrous Synchronization

Cows had their estrous cycles synchronized for first TAI using a Double-Ovsynch protocol starting at 53 ± 3 DIM in herd A and 51 ± 3 DIM in herd B, as described by Souza et al. (2008) and later modified by Brusveen et al. (2009) . The GnRH (100 μg/dose of gonadorelin hydrochloride, Factrel) and the PGF 2α (25 mg/dose of dinoprost tromethamine, Lutalyse) were from Zoetis (Madison, NJ). Briefly, cows received the first GnRH treatment of the Presynch portion of the Double-Ovsynch, followed by a treatment with PGF 2α 7 d later and GnRH 72 h after PGF 2α . Seven days later, cows received a GnRH (G1) treatment followed by 2 PGF 2α treatments administered 7 and 8 d later, with the last GnRH (G2) treatment administered 56 h after the first PGF 2α treatment and AI 16 to 20 h later (Figure 1). Within each herd, cows (n = 800; n = 560 herd A; n = 240 herd B) were blocked by parity (primiparous vs. multiparous) and randomly assigned to serve as untreated controls (high-P4) or to receive a half dose of PGF 2α (low-P4; 12.5 mg of dinoprost tromethamine) 2 d before G1 (Figure 1). At each location, multiple sires with high genetic merit and proven fertility were used and were equally balanced between treatments.
