Dex and corticosterone were prepared in phosphate-buffered saline (PBS). E2 was prepared in ethanol and dissolved in PBS for an experimental concentration of 10 nM.

VCaP cells plated in 6-well dish at confluency of 40-45% were transfected with 30pmol of small interfering RNA (siRNA) against ERG (Dharmacon), AR (Dharmacon), FOXA1 (Dharmacon) and non-targeting control (Dharmacon). Lipofectamine RNAiMAX Transfection Reagent (Thermo-Fisher Scientific) was used according to manufacturer's protocol.
