Twenty micrograms of cell extract was separated by sodium dodecyl sulphate–polyacrylamide gel electrophoresis and electroblotted onto nitrocellulose membranes. Primary antibodies were PHB mouse monoclonal (MS-261-PO; ThermoFisher) against GAPDH (SC-32233 (Santa Cruz, CA, USA); 1:5000) or PHB (SC-28259 (Santa Cruz Biotechnology, Inc., Dallas, TX, USA); 1:1000), MCM5 (SC-22780; Santa Cruz), E2F1 (SC-193; Santa Cruz). Peroxidase-labelled rabbit anti-mouse secondary antibodies (Sigma) were used at 1:2000. The membrane was then incubated in chemiluminescent substrate (GE Healthcare), and light emission detected by autoradiography.
