Volatile Organic Compound Profiling in Fermentation

Volatile organic compound (VOC) profiles were determined following procedures described by Zou et al. [11 (link)], with some modifications. Samples of the FCRB and TFRB were stored at 4 °C before analysis, and 1 µL 2-octanol internal standard and 2 g NaCl were added to each 8 mL sample in a 20 mL vial and tightly capped with a PTFE/silicone septum. Carboxen/polydimethylsiloxane (CAR/PDMS) SPME fibers (75 µm, Fused Silica 24 Ga, Manual Holder, 3 pk, Supelco, Bellefonte, PA, USA) were used for VOC extraction at 60 °C for 50 min. Compounds were desorbed for 5 min at 280 °C in splitless mode, using a 0.75 mm dedicated SPME liner.
Gas chromatography conditions, MS operating conditions, and qualitative and semi-quantitative analysis were all performed according to the method described by Zou et al. [11 (link)].

Solid-Phase Microextraction (SPME) Fiber Selection

The SPME fiber assembly kit assortment, Stableflex 24Ga, manual holder, 4pk fibers and PDMS/DVB (divinylbenzene), fused silica 24 Ga, SPME holder and SPME manifold were supplied by Supelco. The fibers selected were PDMS/DVB and PDMS/DVD+OC (organic carbon) with film thickness diameter 65 µm, polyacrylate 85 µm, Carboxen™/PDMS 85 µm and DVB/CAR/PDMS 50/30 µm. Fibers were conditioned with the set up parameters and tested without further modification. To eliminate potential problems with carry over and baseline noise, blank injections were performed between runs.
