A GAS5 over-expression plasmid, null control, small interfering RNA (siRNA) for GAS5 and negative control (siNC) were designed by GenePharma (Shanghai, China). The siRNA sequences are shown in Table 1 of the Supplementary File , available at Carcinogenesis Online. We stably overexpressed GAS5 in T24 cells by transducing the cells with lentivirus (GenePharma, Shanghai, China). The cells were screened with puromycin for 4 weeks. Stable transfectants were used for animal experiments. The miR-18a-5p mimic, mimic control, miR-18a-5p inhibitor and inhibitor control were purchased from RiboBio (Guangzhou, China). Transfection of T24 and EJ cells was performed with Lipo8000™ transfection reagent (Biyuntian Biotechnology, Shanghai, China). After 48 h of transfection, T24 and EJ cells were collected for further experiments.

For lncRNA CASC2 over-expression, lncRNA CASC2 plasmid was used. The lncRNA CASC2 sequence was synthesized based on the lncRNA CASC2 sequence and then sub-cloned into the pcDNA3.1 vector (lncRNA CASC2-plasmid; Shanghai GeneChem Co., Ltd.). At 48 h after transfection, reverse transcription-quantitative polymerase chain reaction (RT-qPCR) was performed to determine transfection efficiency.

Investigating CASC2 and miR-18a-5p in Melanoma

A375 cells were seeded onto a 60-mm dish and cultured for 24 h. To evaluate the roles of CASC2 and miR-18a-5p on MM, cells were transiently transfected with pc-CASC2, pcDNA3.1, si-CASC2, si-control, miR-18a-5p inhibitor, inhibitor control, miR-18a-5p mimic, mimic control, or pc-RUNX1, respectively (RiboBio Corp., Guangzhou, P.R. China) using Lipofectamine 2000 (Invitrogen, Carlsbad, CA, USA). To further evaluate the relationship of CASC2 and miR-18a-5p, cells were transfected with pc-CASC2, pc-CASC2 + miR-18a-5p mimic, or pc-CASC2 + mimic control, respectively. After treatment for 48 h, cells with various treatments were subjected to the following experiments.
