Detection of Active BAX Protein

For detection of active BAX, cells were lysed in CHAPS lysis buffer (10 mM HEPES (pH 7.4), 150 mM NaCl, 1% CHAPS) as previously described [50 (link)]. Briefly, 1000 μg protein were incubated overnight at 4°C with 5 μg mouse anti-BAX antibody (6A7, Sigma) followed by addition of 10 μL Dynabeads Protein G (Invitrogen) and further incubation for 2h at 4°C. Dynabeads were washed with lysis buffer and BAX levels were analyzed by immunoblotting using rabbit anti-BAX (1:1000; 5023, Cell Signaling).

Briefly, 700-1000 μg protein was immunoprecipitated and incubated overnight at 4°C with 2 μg/ml mouse anti-Bax antibody (6A7, Sigma) or anti-Bak antibody (Ab-1; Calbiochem) and 10 μl pan-mouse IgG Dynabeads (Dako, Hamburg, Germany) and washed with CHAPS buffer. The precipitate was analyzed for Bax and Bak expression by Western blotting using the Bax NT antibody (Merck) or Bak antibody (BD Biosciences).
