Zinc Oxide Nanoparticles Cytotoxicity Evaluation

The TM4 cells were incubated with different concentrations of ZnO NPs (0, 0.2, 0.5, 0.75, 1.0, and 1.25 µg/mL) for 24 and 48 hours. Then, cells were fixed with 4% paraformaldehyde for 20 minutes at room temperature. Fixed cells were blocked with 0.1% Triton X-100 (Merck, USA) and 3% BSA in PBS for 30 minutes after permeabilizing by 0.1% Triton X-100 in Triton/PBS solution for 15 minutes. Subsequently, cells were incubated with rabbit anti-LC3B (1:100) (Thermo Fisher Scientific, USA), mouse anti-Arp3 (1:50) (Sigma-Aldrich, USA), mouse anti-α-tubulin (1:300) (Abcam, USA), and rabbit γ-H2AX (1:200) (Millipore-Sigma, Burlington, MA, USA) in 0.1% TX-100/PBS overnight at 4 °C. After washing twice with 0.05% Tween-20/PBS, cells were incubated with goat anti-mouse Dylight 650 (1:1,000) (Thermo Fisher Scientific, USA), goat anti-rabbit Dylight 550 (1:1,000) (Thermo Fisher Scientific, USA) and the DNA staining dye Hoechst 33342 (Molecular Probes, Eugene, OR, USA) in PBS/BSA for 90 minutes at room temperature in the dark, followed by incubation with 50 µL Alexa Fluor 488 Phalloidin (Cell Signaling Technology, Danvers, MA, USA) per well at room temperature for 30 minutes for F-actin staining.
