Apoptosis and Cell Cycle Analysis in HCT116 PTPRS KO Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
HCT116 PTPRS KO Cells and control cells were treated with the ERK inhibitors SCH772984 (942183-80-4 Santa Cruz Biotechnology), VX11-E (A3931 ApexBio), VRT752271 (B1106 ApexBio); MEK inhibitors Trametinib (A3018 ApexBio) and AZD8330 (A8374 ApexBio); PI3K inhibitor LY294002 (Cell Signaling 9901), Dasatinib (CDS023389 Sigma) and the AKT inhibitor VIII (A6730 Sigma). All treatments were for 48 hours and the noted concentrations, unless specified otherwise. Following treatments, Annexin 5 apoptosis assay or a cell cycle analysis using Brdu and propidium iodide staining were performed as described previously [42 (link)]. The analyses were performed with a BD Accuri-C6 Flow Cytometer.

Inhibitors Screening in Breast Cancer Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
MCF7, MDA-MB-231, and SUM-159 cells were cultured in Dulbecco’s Modified Eagle’s Medium (Hyclone, Logan, UT, USA) supplemented with 10% fetal bovine serum (FBS), 100 U/mL penicillin, and 100 mg/mL streptomycin. All cell lines were cultured in a humidified atmosphere of 5% CO2 at 37 °C. For treatments with the following inhibitors, cells were seeded at the same density and allowed to adhere for 24 h under normal conditions before treatment with the indicated concentrations of different inhibitors. The inhibitors including AZD8330 (A8374), SB203580 (A8254), SP600125 (A4604), PS-1145 (B6089), (5Z)-7-Oxozeaenol (B7443), MK2206 (A3010), LY2109761 (A8464), and Cyclo-RGDfK (A8164) were purchased from APExBIO (Houston, TX, USA). Belvarafenib (S8853) was purchased from Selleck Chemicals (Houston, TX, USA), and UK122 (sc-356185) was purchased from Santa Cruz Biotechnology (Santa Cruz, CA, USA).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
