Cloning and Antibody Production for LGTV EDIII

Check if the same lab product or an alternative is used in the 5 most similar protocols
The LGTV Envelope domain EDIII gene was amplified from nymphs’ cDNA and cloned into the pET-28a(+) expression vector [24 (link)]. A rabbit polyclonal anti-EDIII antibody was generated against recombinant EDIII protein (rec EDIII) in GL Biochem, Shanghai, China [25 (link)]. To determine the specificity of the anti-LGTV antibody, E protein levels were compared between ticks and cells infected with or without LGTV. Mouse anti-β-Actin antibody (1:2000) (Abbkine, Wuhan, China) was used as an internal control.

Immunoblotting of Midgut Proteins

Check if the same lab product or an alternative is used in the 5 most similar protocols
Proteins of 10 midguts 24 h post blood meal were extracted in 300 µl SDS/urea lysis buffer (8 M urea, 2 % SDS, 5 % β-mercaptoethanol, 125 mM Tris-HCl).
Immunoblotting was performed using standard procedures using the antibodies rabbit anti-Per1 antibody (1:5000) and mouse anti-β-Actin antibody (1:2000)
(Abbkine, China). The rabbit polyclonal anti-Per1 antibody was generated against recombinant Per1 protein (recPer1) corresponding to bases 55-462 of per1 CDS (Aste010406) expressed in pET-42a (Novagen) commercially (GL Biochem Ltd, Shanghai, China).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
