Neutrophil viability assay with Sinomenine

Neutrophils isolated from the peritoneal cells of mice were inoculated into 96-well plates (100 μL/well, 5 × 10⁶ cells/well) with or without different concentrations of Sinomenine. The neutrophils were cultured at 37 °C in a 5% CO₂ incubator. After four hours of culture, Cell Counting Kit 8 (CCK-8) solution (10 μL/well) was added to each well and incubated for 1 h. The absorbance at 450 nm was measured with a microplate reader (BioTek Instruments, Inc).
