Single cell splenocyte suspensions (8 × 10⁵ cells/well) were cultured in U-bottom culture plates (Greiner, Frickenhausen, Germany) with either medium or OVA (50 µg/mL) for four days at 37 °C, 5% CO₂. Culture supernatant was collected and stored at −20 °C until measurements of IFNγ, IL-13 and IL-10 by means of ELISA, as described elsewhere [17 (link)].
