Cytokine and Chemokine Profiling After BCG Immunization

Sera were collected from LMC and M-Traf3^-/- mice at day 1 or day 2 after the 2^nd and 3^rd BCG injections. Serum levels of 40 cytokines and chemokines were measured using a Mouse Cytokine Array Panel A kit (R&D, Minneapolis, MN) following the manufacturer’s instructions (27 (link)). Images of the blots were acquired and quantitative analyses of cytokine/chemokine spots were performed using a low-light imaging system (LAS-4000 mini, FUJIFILM Medical Systems USA, Inc., Stamford, CT) (27 (link)). Concentrations of G-CSF, CXCL-13, S100A8/A9, HMGB1, and CCL22 in mouse sera were determined by quantitative ELISA using specific coating Abs and biotinylated detection Abs (R&D or Biomatik) following the manufacturer’s protocols.
