Establishing Lymphoma and Melanoma Cell Lines

For in vitro experiments, the Burkitt lymphoma cell line Raji as well as the melanoma cell line 526-Mel were transduced with lentiviral vectors encoding a GFP-firefly luciferase cassette. GFP⁺ 526-Mel cells were additionally modified to stably express CD20. For in vivo studies, a mouse-adapted Raji^ffLuc cell line was used [37 (link)]. All tumor cell lines were cultured in RPMI 1640 (Biowest, Nuaillé, France) supplemented with 2 mM glutamine (Lonza, Basel, Switzerland) and 10% fetal bovine serum (FBS, Biochrome, Berlin, Germany). The human embryonic kidney cell line 293 T was cultured in Dulbecco’s modified Eagle medium (Biowest, Nuaillé, France) supplemented with 10% FBS.
