Twenty weeks after humanization, 12 Hu-mice were infected intraperitoneally (IP) with 2 × 104 tissue culture infectious dose 50 (TCID50) of HIV-1ADA (a macrophage tropic viral strain) (61 (link)–63 (link)). An additional six Hu-mice served as uninfected aged-matched controls. Peripheral blood samples were collected every four weeks via submandibular vein bleeding to assess HIV-1 viral RNA and to assess the dynamics of human immune cell markers by flow cytometric analysis. Plasma HIV-1 RNA levels were measured using an automated COBAS Ampliprep V2.0/Taqman-48 system (Roche Molecular Diagnostics, Basel, Switzerland) as per the manufacturer's instructions. The detection limit after dilution factor adjustment was 200 viral RNA copies/mL.
