Highly Purified Platelet Isolation

Platelets were isolated and purified via density centrifugation and immune depletion of CD45⁺ cells using human CD45 MicroBeads reagent (Miltenyi Biotec GmbH, Bergisch Gladbach, Germany) according to manufacturer’s instructions. [15 (link),16 (link)]. At last, a MACS^® bead separation system (Miltenyi Biotec GmbH) was used to collect leukocyte-depleted platelets with purity >99.99%. Platelet RNA was extracted RNA with AG RNAex pro (Accurate Biotechnology (Hunan) Co., Ltd., Changsha, China). An Agilent Bioanalyzer was used for RNA quality control.
